Preparation Of Compond Metronidazole Effervescent Tablets And Study On Its Pharmacodynamics

Endometritis is the mucinous or pyogenic inflammation of the womb mucosa membrane. As a common reproductive organ disease especially for cows,it creates great losses to the cow industry. Although the pathogenic microorganisms causing the inflammation are many and varied,pathogenic bacteria are mostly factor,including aerobes and anaerobes,which may create infection to the womb under suitable conditions.Metronidazole is one of the main medicines for anti- anaerobes clinically. In order to expand its antibacterial spectrum and facilitate the clinical practice,the prescription of the compound metronidazole effervescent tablet was designed with metronidazole , oxytetracycline hydrochloride,and other chemical medicines. Effervescent tablet,a quite novel tablet dosage-form,was chosen for the preparation dosage-form so as to increase a new preparation for clinical treatment to the cow endometritis.1. Establishment of prescription and craft Through the adjuncts choices and the orthogonal experiment,the prescription of the compound metronidazole effervescent tablet has been established: metronidazole 0.2g,oxytetracycline hydrochloride 0.25g,and other chemical medicines. The particles of adjuncts were made with absolute ethyl alcohol,and the acid and alkali were dried respectively to prepare the tablets with the particles together.2. Quality control The quality examinations of the compound Metronidazole effervescent tablet indicated that the tablets conformed to the pharmacopoeia requests: it was completely broken up in 5 min,and the average foaming quantity reached 35.6 mL. The Contents of metronidazole and oxytetracycline hydrochloride were determined respectively by dual wavelength ultraviolet spectrophotometry and single wavelength ultraviolet spectrophotometry,and the results showed that the average recoveries were respectively 99.49% and 100.76%,which indicated that the determining method was accurate and reliable. The stability tests were done to estimate the period of validity of the compound metronidazole effervescent tablet,which was approximate 1 year.3. Pharmacodynamics study The pharmacodynamics of the compound metronidazole effervescent tablet was inspected by anti-bacterium experiment in vitro and the clinical experiment. The MICs to the test bacteria were all below 6.25μg·mL^-1. The clinical experiment result indicated that the compound metronidazole effervescent tablet efficiency for treating the cow endometritis was 94.12%.Conclusions: The preparation craft of the compound metronidazole effervescent tablet is simple,and the quality indexes completely conform to the pharmacopoeia,especially the huge foaming quantity showes that its effervescent performance is superior. The compound metronidazole effervescent tablet is quite sensitive to the environment. the compound metronidazole effervescent tablet expands the antibacterial spectrum and is suitable to treat endometritis.
