Herba cirsii is the whole grass of Cirsium setosum(Wild.)MB. which is a kind of perennial herbaceous plant, belongs to the family of Compositae. As a species of the genus, Herba cirsii is also called Mao-ji, Qing-ci-ji, Qian-zhen-cao, etc. Herba cirsii has extensively distributed in all over the China, and is very rich in the plant resources in Heilongjiang. It was recorded originally in《MingYiBieLu》and has been recorded in every edition of《Chinese Pharmacopoeia》including the newest edition in 2005. As the traditional Chinese medicine which has a long history to be used, it can cool the blood and stop bleeding, and other pharmacological activities. Nowdays, It has been reported that clinical use of Herba cirsii showed significant effect on the treatment of glomerulonephritis (GN). However, few of works on its Pharmacodynamic Substance Basis related to the treatment of those diseases are reported . Chemical and pharmaceutical research have been conducted on the part extracted with EtOAc from for the aims of searching the effective section and further determining the active compounds for hemostasis and antiinflammatory.At first, the experiments of chemical and pharmacodynamics were performed on the bioactive fraction from the extracts of the Herba cirsii. The results confirmed that the extract with EtOAc vary the actions on stopping hemorrhage, hemostasis and antiinflammatory. It is the first time that the chemical constituents extracted with EtOAc from Herba cirsii was confirmed to be the effective part for hemostasis and antiinflammatory.In the course of our studies on chemical constituents in effective parts of hemostasis and anti-inflammatory, 15 compounds were isolated with silica. gel, reversed-phase(ODS) column chromatography and HPLC, of which the structures of 6 compounds were fully determined according to the chemical and physicochemical evidence (1Dand2D-NMR). 4 compounds were firstly found in Cirsium setosum(Wild.)MB.Among them there are 4 flavonoids compounds and 2 lignans compounds. This thesis further confirmed the pharmacological action of effective part in Herba cirsii and the effective substances for hemostasis and antiinflammatory. All these will provide important scientific foundation for further research and the development of new drugs and will promote the study and understanding of the mechanism of Herba cirsii against GN.
