Background:The Cirsii Japonici Herba Carbonisata is a dried aerial part of Cirsium japonicum DC.It can cool blood,stop bleeding,and reduce swelling.It is widely used throughout the country.In clinical practice,Cirsii Japonici Herba Carbonisata is commonly used to treat hemorrhagic diseases.Studies have shown that the effect of hemostasis after the charcoal is enhanced.Some studies have suggested that phylloxera is a hemostatic component.However,due to the decreased content of melittin in the component willow after the charcoal decoction,the hemostatic effect is enhanced,and there are also some conclusions about carbon and other effects on hemorrhage.However,it is a contradiction.What is the hemostatic component of Cirsii Japonici Herba Carbonisata?In order to solve this problem,we used the research methods of modern nanomaterials,and carried out the following experimental research on Cirsii Japonici Herba Carbonisata.Expected to reveal the mystery of the hemostatic.Objective:In this study,the characterization technology of nanosciences was introduced.Cirsii Japonici Herba Carbonisata was used as the research object to explore the material basis for the enhanced hemostasis after chaarcoal.From the perspective of nano-science research,the carbon-dots in Cirsii Japonici Herba Carbonisata were analyzed,and the hemostatic effect and mechanism of Cirsii Japonici Herba Carbonisata were analyzed from the perspective of pharmacology and pharmacodynamics.Therefore,it was confirmed that the Cirsii Japonici Herba Carbonisata carbon dots in Cirsii Japonici Herba Carbonisata,thus revealing the hemostasis component of Cirsii Japonici Herba Carbonisata.Methods:(1)Preparation of nano-components in Cirsii Japonici Herba Carbonisata:The use of a muffle furnace to burn large-scale carbons at different temperatures,and the nano-materials were used to separate the components of the carbon from Cirsii Japonici Herba Carbonisata to nano-materials.Structure identification methods characterize the components in the charcoal fired at different temperatures and use the pharamacodynamics research method to optimize the temperature.Provide the cornerstone for the next study.(2)Comparative study of nano-components of Cirsii Japonici Herba and Cirsii Japonici Herba:In order to relieve the trouble of the hemostasis component in big cockroaches as a small molecule,the nano-components in the separated cockroach charcoal were compared with those in the cockroach.Using high performance liquid chromatography(HPLC),a simple comparison of the compositional differences in the large cockroaches and big cockroaches was conducted to analyze the differences in the components of the purified cockroach and cockroach.(3)Characterization of Nano-sized Components in Cirsii Japonici Herba Carbonisata:Using the research techniques and methods of nano-materials direction,the purified big cocoon carbon composition was identified,and UV-visible spectrophotometer(UV-vis)and Fourier infrared were used.Spectroscopy(FT-IR),X-ray photoelectron spectroscopy(XPS),high-resolution transmission electron microscopy(HR-TEM)and other commonly used analysis methods in the field of Carbonisata.According to the literature study,the appropriate bleeding model was selected:the tail bleeding and hepatic hemorrhage model.To study the effect of nano-components in Cirsii Japonici Herba Carbonisata on the bleeding time of mice with different bleeding models,and to analyze the strength of its hemostatic effect.(5)Study on the mechanism of hemostasis of Cirsii Japonici Herba Carbonisata carbon nano-components:explore the mechanism of hemostasis of nano-components in giant salamander and evaluate its effect on the coagulation system by measuring four coagulation factors in animals;determine the platelet count of experimental animals.The number and platelet activity were evaluated in order to evaluate its effect on platelets;factors such as t-PA in mice administered were measured to evaluate the effects of nano-components in Cirsii Japonici Herba Carbonisata on the fibrinolytic system in mice.Through this series of studies,the hemostatic effect of nano-components in Cirsii Japonici Herba Carbonisata was demonstrated,and its hemostatic mechanism was studied.Results:(1)Preparation and optimization of conditions for nano-components in Cirsii Japonici Herba Carbonisata:This experiment was used to prepare large-scale alfalfa charcoal with different temperatures,and it was isolated and characterized.Under the electron microscope,the morphological characteristics of the big cocoon carbon purification solution at different temperatures are different,mainly in terms of the size distribution of the particle size;and infrared and other spectroscopic studies have also found that the group types at different temperatures are significantly different.It was found that the charcoal fired at 350? has more similar nanomaterial characteristics in morphology and group type.Through the study of the components extracted from the big cockroach charcoal fired at different temperatures,it was found that the components fired at 350?.had a better hemostatic effect,so a firing temperature of 350?.was selected.(2)Comparative study of nano-components of Cirsii Japonici Herba Carbonisata and Cirsii Japonici Herba Carbonisata carbon dots:In this experiment,the differences in the components extracted from Cirsii Japonici Herba Carbonisata and Cirsii Japonici Herba Carbonisata carbon dots were analyzed by high-performance liquid chromatography.After the carbonization of Cirsii Japonici Herba Carbonisata,the small molecule components were not shown in the HPLC results after dialysis to remove the small molecule components,while the components in the large scale were complicated under the same conditions.(3)Characterization of Nano-sized Components in Cirsii Japonici Herba Carbonisata:The analysis of Cirsii Japonici Herba Carbonisata dialysate by UV-Vis spectrophotometer,Fourier transform infrared spectroscopy,and projection electron microscopy showed that:Shows evenly distributed ball species with a particle size distribution between 2-11 nm with a lattice spacing of 0.297.IR results show that the substance is rich in radicals,with groups such as-OH,-COOH,and UV-visible spectroscopy.The photometer shows that it has weaker UV absorption.(4)Study on the hemostatic effect of nano-components in Cirsii Japonici Herba Carbonisata:The hemorrhagic model of mice was administered in vivo.The results showed that Daqi carbon dialysate had the effect of prolonging the bleeding time of mice.The results showed that for various animal hemorrhage models,Cirsii Japonici Herba Carbonisata carbon dialysate showed good hemostatic effect.(5)Study on the mechanism of hemostasis of Cirsii Japonici Herba Carbonisata carbon nano-components:Effect on the coagulation system The experimental results show that daxue carbon extract neither affects the exogenous coagulation system nor the endogenous coagulation system,but it does not affect the FIB.Has an elevated effect and is superior to the positive drug group.Through the platelet number and platelets are affected,it may be through the impact of platelets exert their hemostatic effect.At the same time,the study of the kit showed the effect on the fibrinolytic system.Conclusion:In this experiment,a method of nanomaterials was used to separate the nanosized carbon components from Cirsii Japonici Herba Carbonisata,and the appropriate conditions were optimized.Through different mouse models,the nano-components in Cirsii Japonici Herba Carbonisata were The efficacy of the drug is discussed.To study the effects of Cirsii Japonici Herba Carbonisata nano-components on mouse tail-tip hemorrhage model and hepatic hemorrhage model;and to determine four coagulation factors in rats,and to study the effect of nano-components of Cirsii Japonici Herba Carbonisata on the blood coagulation system;Effects of Carbon-rich Quantum Dots on Fibrinolytic System.This study provides experimental evidence and guidance for the basic research on the quality of Chinese herbal medicines.The material basis research of traditional Chinese medicines is of great significance,but the current experimental methods and methods have not been given a reasonable explanation.This experiment took advantage of the research methods and means of nanomaterials to explore the material basis of the hemostasis effect of Cirsii Japonici Herba Carbonisata,and to provide guidance and methods for the study of hemostatic components of Chinese traditional medicine.
