Objective:To investigate the effect of ginseng total saponins (GTS) on nerve growth factor (NGF) protein expression in polycystic ovary (PCO) rats,and the influence of testosterone and androstenedione in serum.Methods:Rats received IM injection of estradiol valerate (EV) to induce PCO,drug dose and action time were defined.Then,other rats received the exact dose to result in PCO and administered GTS intraperitoneally every other day beginning 1 day after the EV injection.Observe the NGF protein expression in the hypothalamus,pituitary,ovary and adrenal,and determine the serum testosterone and androstenedione.Results:1) Polycystic ovaries were fully developed in rats with a single intramuscular injection of 4mg EV after 30 days.2) The level of serum testosterone and androstenedione in PCO rats was not obvious advanced compared to the normal.Increased expression of NGF was noted in the ovaries and the brain of rats with PCO.3) GTS administration attenuated NGF expression in the ovaries but not in the brain.Conclusions:It is the most typical polycystic ovary model after rats received 4mg EV 30 days.In this model,there was no obvious advance compared to the normal of the serum testosterone and androstenedione.The NGF protein expression strengthen then the normal in ovary and brain tissue.And GTS can cut down the excess expression of NGF,but there was no alternation in the brain tissue.
