Objective To observe the changes of bone density, blood rheology, ineraossonspressure in rats of diabetes model, and analyze their roles in the pathologcal changes ofavascular necrosis of the femoral head induced by diabetes.Methods Totally 40 rats of 12 weeks old were used, 14 rats(7 males and 7 females)were selected at 2 weeks after adaptive feed as controls(control group). The other 26 ratswere made into models of immediate streptozotocin-diabetes mellitus by usingintraperitoneal injection of streptozotocin with the dose of 60mg/kg. And then the modelswere injected with long-acting insulin(4U/kg) from the 24th hour after modelestablishment once every other day, which was taken as basic insulin to maintain life.After the model establishment, sufficient water and feed were given, uric glucose wasdetected once everyday. Blood samples were dawn from tail to detect the nonfastingblood glucose at 48hours and 1 week respectively. The rats whose uric glucose was stillabove +++ and twice blood glucose both above 16.7mmol/L were taken as thesuccessfully established diabetes mellitus, and 14 of them (7 males and 7 females) wereselected as the diabetes group. The rats in the control group were injected with saline ofthe same volume. The changes of bone density, blood rheology, ineraossons pressure wererespectively detected at 12 weeks.Results Of the 40 rats, 28 were involved in the analysis of results. 1.The bonedensity in the diabetes groups was 0.183±0.047g·cm-2, while in the control group was0.192±0.094g·cm-2. There was singnificant difference between the two groups(P<0.05).2. The lower shear rates, higher shear rates, plasma viscosity, RBC aggregate andhematocrit of diabetes groups were respectively 14.93±1.37mpa·s, 5.78±0.49mpa·s, 1.32±0.09mpa·s,2.58±0.21 and 53.55±2.94. And in the control group these wererespectively 17.27±1.40mpa·s,6.18±0.53mpa·s,1.40±0.08mpa·s,2.83±0.25 and59.29±3.343. Comparing the two groups, there was singnificant difference(P<0.05). Theinera- ossons pressure was respectively 86±8mmHg and 40±5mmHg in the diabetesgroups and control group. There was singnificant difference between the twogroups(P<0.05). defieciency of insulin leads to bone density increased, ineraossons pressure depressed andblood theology changed. All these accord with the pathological changes in necrosis of thefemoral head. So this is an important pathological process and deadly cause for diabeticavascular necrosis of the femoral head.
