ObjectiveTo investigate the effect of 2, 3-dioxoindoline on apoptosis and human telomerasereverse transcriptase mRNA in SH-SY5Y cells.MethodsChange of nucleolus stained with Hochest33258 was observed with fluoroscope; DNAfragment was observed with agarose gel electrophoresis; the expressions of bcl-2 andbax protein were analyzed by Western blotting; the hTERT mRNA level was examinedby RT-PCR.ResultsAfter cells were treated with ISA of different concentrations (0,100,200,400μmol·L-1)for 48 h, Hochest33258 staining could distinguish apoptotic cells as well as normal cells.DNA fragment was found in cells treated with 200μmol·L-1 and 400μmol·L-1 ISA byagarose gel electrophoresis. The expression of bcl-2 protein was decreased, as comparedwith that of the control (P<0.05). However, the expression of bax protein kept constant(P>0.05); and the expression level of hTERT mRNA, the key subunit of telomerase,decreased after 2, 3-dioxoindoline treatment compared with that of the control (P<0.05).Conclusion2, 3-dioxoindoline induces apoptosis of SH-SY5Y cells in dose-dependent manner. Thepossible mechanism was due to the decrease of the level of bcl-2 and hTERT.
