Objective: In this study, we established a rat model of small intestinal ischemicpreconditioning to see if there is protective effect against acute myocardialinfarction, and to study the relativity between the protective effect and the timeof small intestinal ischemic preconditioning. To assess if the mechanism of theprotective effect is related to the change of AngⅡand mRNA expressions of AT1acceptors and PKC in rats.Methods: The rats were randomly distributed into five groups: A,B,C groups createdsmall intestinal ischemic preconditioning model with the different times, D groupcreated sham-operated group, E group created control group. After 24 hours, allgroups created the model of acute myocardial infraction in rats. Then after 24 hours,the model succeed, the cardiac musclar tissue and the blood serum were taken. Theischemic and myocardial infarct size and creatinekinase-MB in serum were measuredto assess the protective effect. And the conceivable mechanism is determined by thecontent of AngⅡ, protein kinase C and the mRNA content of AT1 acceptor in myocardiumtissue. The mRNA content in AT1 acceptor is detected by the method of RT-PCR.Results: In the group of small intestinal preconditioning, compare with othergroups, the ischemic and myocardial infarct size and creatinekinase-MB in serum ismarkedly reduced (P<0.05), the content of AngⅡand the mRNA content of AT1 acceptorin myocardium tissue are also obviously decreased (P<0.05), the content of proteinkinase C is increased (P<0.05). These trends relate to the time of small intestinalischemic preconditioning.Conclusions: Small intestinal preconditioning could reduce cardiac infarct size,lessen the damage of myocardium. And the protective effect is related to the timeof small intestinal ischemic preconditioning.
