ObjectiveThis study chose the factor COX-2 that are associated with the angiogenesis. Detection of the expression of COX-2 in vulvar diseases, and to analyze its relationship with microvascular density. COX-2 is to discuss the changing relationship with squamous cell carcinoma and the relationship with angiogenesis.MethodsMaterials: 30 cases of vulvar squamous carcinoma; 35 cases VIN (20 VINⅠ-Ⅱ, 15 VINⅢ); 40 cases of NNEDV (20 squmous cell hyperplasia,20 lichen sclerous) and 10 cases of normal vulvar skin from January 1995 to January 2005 the First Affiliated Hospital of China Medical University pathology office filed after surgical removal and biopsy specimens. Reagents: COX-2 polyclonal rabbit anti-human antibody, the concentration of 1:100, rabbit polyclonal anti-CD34 antibody, work concentration 1:80. Using immunohistochemical staining method of SABC. T-test to differentiate the rates of different groups for the expression of COX-2 in vulvar diseases and MVD were analysed.If (P<0.05) having significant differences,SPSS version 13.0 software was employed to analyze all data.Results1,The expression of COX-2: The expression of COX-2 protein in the nucleus and cytoplasm, is brown granules. In NNEDV and VIN are expressed in the superficial dermis, and the pathways, COX-2 protein expression in tumor necrosis and tumor-infiltrating the region to the brink of a cancer nest expression. COX-2 is not expressed in normal vulvar skin. The expression of COX-2 in vulvar squamous cell hyperplasia is 3.58±0.421. Both were significantly different (P<0.05); The expression of COX-2 in the vulvar lichen sclerosis is 5.11±0.595. There is a significant difference with squamous cell hyperplasia (P<0.05). The expression of COX-2 in VINⅠ-Ⅱand VINⅢare 4.32±0.544 and 9.63±1.309 respectively, a significant difference was found between the two groups (P<0.05).2,The expression of MVD: The MVD in the tissues of lichen sclerosis was 10.34±2.833,there is significant differences compared with normal vulvar skin (P<0.05)and the normal vulvar skin is not significant differences compared with squmous cell hyperplasia(P>0.05). In the tissues of squmous cell hyperplasia was 21.58±2.475, there was significant differences compared with lichen sclerosis(P<0.05); MVD in vulvar squamous carcinoma was 39.88±7.650. and there is significant difference between VIN(P<0.05). The expression of VTNⅠ-Ⅱand VINⅢwas significant difference(P<0.05).3,The realation between COX-2 and clinical and pathological features of vulvar carcinoma: The expression of COX-2 in the group of lymph node metastasis was significantly higher than that in those without lymph node metastasis (P<0.05), there are no significant differences in different age groups,clinical stage and grade (P>0.05).4,The relation between COX-2 and MVD: The MVD of 15 cases with COX-2-positive is significantly higher than that of the 15 cases of COX-2-negative, suggesting that COX-2 stimulates neoplasia through increased angiogenesis.Conclusion1,COX-2 is not the main factor of. invalid tissue inflationary reflection and micro-angiogenesis of LS and SH.2,COX-2 has the effect of stimulating angiogenesis in dermis of VIN.3,COX-2 can promote angiogenesis and increase the activity of tumor invasion and metastasis in VSCC.
