Objective:To confirm the hypolipidemic therapy effect of XIEZHIPING whichcomposed of Alisma Rhizoma the principal drug and probe the mechanismof hypolipidemic action from the view of animal model experiment in vivoto cell culture in vitro, and finally establish the base of new naturalhypolipidemic product by the way of experiment on the golden hamsterhyperlipoidemia animal model and HepG2 cell line culture.Methods:1. Theoretical research: To review respectively the theoreticaladvance of hyperlipoidemia from both points of TCM and modern westmedicine. Firstly, discuss detailed and completely the part of TCMincluding etiological factor, pathogenesis, clinical classification andtherapies, choosing of prescription and medication; Secondly, comment thepart of modern medicine from etiological factor, pathogenesis, diagnosisto measures of therapy;Thirdly, analyse the background of the compositionof XIEZHIPING, which origins from the clinic application and effect,keeps in accordance with the principle of prescription should coincidencewith syndrome of TCM, also supported by chemical composition of medica materia and modern pharmacology.2. Experimental research:①experiment in vivo: chosen golden hamsteras hyperlipoidemia animal model, after fed 30 days with high-fat-food,detected the items of TC,TG,HDL-C,LDL-C to judge the state ofpathological model. Then given various dosage (high, media, low) ofXIEZHIPING for 30d, detected the 4 items of blood-fat again and slicedliver pathological section to disclose the effect of therapy.②experiment in vitro: chosen HepG2 cell line as the cell model, afterdetermined the safe experimental dosage by cytotoxicity essay of rawextract of XIEZHIPING, add the different dose of extract into the culturedcells to measure the content of cholesterol in the cell lysate andcultured meium with the Amplex(?) cholesterol kit.Results:1. Experiment in vivo:①Weight gain after model made: After 30 daysfed with high-fat-food, the weight of high-fat-food groups increasedremarkably higher than that of common-food group (P<0.01), however, therewas no obvious variance between each high-fat-food group (P>0.05).②changes of 4 items of blood-fat after model made: After 30 days fedwith high-fat-food, the values of TC,TG,LDL-C in high-fat-food groupswere higher than that in common-food group (P<0.01), while the valueof HDL-C decreased obviously(P<0.01); and there was no obvious variancebetween each high-fat-food group of 4 items of blood-fat.③changes ofthe 4 items of blood-fat after XIEZHIPING treated: XIEZHIPING treatedgroups compared with model group after 30 days' administered, the valueof TC,TG,LDL-C decreased remarkably(P<0.05, P<0.01), HDL-C increasedgreatly (P<0.01).④Affect on the liver pathological state afterXIEZHIPING treated: the golden hamster hepatic tissue morphous structureof normal control was normal, which had regular hepatic lobules and nolipid droplet: that of model control manifested diffuse heavy adiposedegeneration and heavy ballooning degeneration with inflammatory cell infiltrate which mainly composed of lympholeukocyte; that of high dosegroup showed midrange ballooning degeneration with chronic inflammatorycell infiltrate which mainly composed of lympholeukocyte and plasmocyte;that of media dose group indicated slight focus ballooning degenerationwith a small quantity normal hepatic cells in some locals; that of lowdose group appeared midrange adipose degeneration with a fewlympholeukocyte cell infiltrate in the parts of central veins and portarea.2. Experiment in vitro: It appeared cytotoxicity when theconcentration of crude extract of XIEZHIPING reached 5mg/ml. Finally,the experimental concentration was arranged as 0,0.25,0.5,1mg/ml afterpreliminary test, the result showed that the crude extract couldstrikingly decreased the content of cholesterol in cell lysate and therewas a positive dose-effect relationship which the content of cholesterolwent down as the concentration went up. The potency of loweringcholesterol of 1mg/ml crude extract of XIEZHIPING was equal to 3μMLipitor; while there showed no affects on the amount of cholesterol inthe cultured medium. The results suggested that the functional mechanismof XIEZHIPING may be that it inhibits the synthesis of cholesterol inHepG2 cell and leads to hypolipidemic effect. However, it has littleimpact on the conveying and excretion of cholesterol from the internalto external of HepG2 cell. The result of experiment in vitro confirmedthe function of lowering cholesterol which was in coincidence with thatin vivo.Conclusions:1. XIEZHIPING can decreased remarkably the value of TC,TG,LDL-Cin animal model of golden hamster (P<0.01, P<0.05), while increasedgreatly the value of HDL-C(P<0.01).2. XIEZHIPING can recover the ballooning degeneration of hepatictissue in model golden hamster. 3. Cude extract of XIEZHIPING through inhibiting the synthesis ofcholesterol in HepG2 cell and leads to hypolipidemic effect.4. XIEZHIPING can treat Hyperlipoidemia with Syndrome of Phlegm andWet obstruction.5. The method of damp elimination, invigorate the spleen and dissipatephlegm, regulating vital energy is effective and feasible in the therapyof hyperlipemia.
