AIM:To investigate the effects of Purple Common Perilla Extract (PCPE) on proliferation and differentiation of HaCaT cells, and then provide foundation to exploit new therapy for some related dematologic disease.METHODS:1.PCPE was extracted by physical measures at common temperature;2.The OD value of living cells was measured by MTT test;3.The surface marker of cells differentiation (CD147) was determined using flow cytometry.RESULTS:1.Microscopy shows: After being interfered by PCPE, the amount of HaCaT cells was tendency to increase as the concentration increased, and the shape of cells became thin flat and vague, just appear to be well-differentiated compared with control group.2.MTT results show: the proliferation of HaCaT Cells was significantly inhibited (P<0.05) after being interfered by PCPE for 24 hours when contrast to the control group, and the original effective concentration was 60g/L. But after 48 hours and 72 hours reaction, the proliferation of HaCaT cells could be significantly inhibited (P<0.05) at each concentration of PCPE, and the inhibitory effect was enhanced as the increase of PCPE's concentration that means it appeared to be dosage-dependent(P<0.01); the best effective time was at 48 hour compared with at 24 hour and at 72 hour(P<0.05).3.FCM results show: The total value of fluorescence which reflected the intensity of protein CD147 on the surface of HaCaT cells was significantly reduced (P<0.01) after being interfered by PCPE for 48 hours when contrast to the control group, and the original effective concentration was 20g/L .The value was reduced as the increase of PCPE's concentration, that means it appeared to be dosage-dependent (P<0.01).CONCLUSION:PCPE has inhibitory effect on proliferation and inductive effect on differentiation of HaCaT cells in vitro.
