Effects Of Splencectomy On The Expression Of TGFβ1 In The Liver And The Serum Level Of TGFβ1 Of The Rats With Hepatic Fibrosis

Objectives: TGFβ1 is one of important factors which can promote the development of hepatic fibrosis. To discuss the effects of splenectomy on hepaic firosis, we tried to study the effects of splenenctomy on the expression of TGFβ1 in the liver and the serum level of TGFβ1 of the rats with hepatic firosis.Methods: By hypodermic injection CCL4,we established 50 rat models with hepatic fibrosis,then took different samples of the livers and spleens of the rats in 3W,6W ,and 8w. We also took liver samples of the rats with splenectomy after 4W.The expressions of TGFβ1 in the spleen and liver were detected by immunohistochemistry technique and the degree of hepatic fibrosis were detected by HE and Masson's staining. IOD for TGFβ1 were measured by CMIAS8 computer colour image analysis systems. The serum levels of TGFβ1 were analyzed by ELISA technique.Results:(1) TGFβ1 was mainly distributed not only in cytoplasm of hepatic interstitial cells that lay in portal areas fiberseptation inflammation areas, but also in a few hepatocyte cytoplasm and a little in fibrous tissue and normal tissue. IOD for TGFβ1 in the liver in different periods were: 26.81±5.42 (normal),80.21±10.98 (3W), 134.60±12.12(6W), 204.60±15.76 (8W). The results indicated that with the development of hepatic fibrosis ,the expression of TGFβ1 in the liver increased (P<0.01).(2) In the spleen, TGFβ1 was mainly distributed in cytoplasm of splenic interstitial cells and a little in fibrous tissue and normal tissue. IOD for TGFβ1 in the spleen in different periods were: 22.40±4.76(normal),59.96±10.17(3W), 98.81±7.40(6 W), 180.46±11.03 (8W).The results also indicated that with the development of hepatic fibrosis ,the expression of TGFβ1 in the spleen increased (P<0.01).(3) The serum levels of TGFβ1 before splenectomy ,2W and 4W after splenectomy respectively were (pg/ml): 380.565±35.826, 394.149±36.297, 417.087db31.785. The serum levels of TGFβ1 of the rats without splenectomy in the same periods respectively were (pg/ml): 385.870±35.359, 421.014±38.663, 442.808±39.228.The results indicated that the serum levels of TGFβ1 of the rats after splenectomy were significantly lower than which of the rats without splenectomy (P<0.05 ) .(4) IOD for TGFβ1 in the liver of the rats with splenectomy after 4W was 224.35±9.45,and IOD for TGFβ1 in the liver of the rats without splenectomy in the same period was 240.40±11.73. The expressions of TGFβ1 in the liver of the rats with splenectomy were less than which of the rats without splenectomy (P<0.05) .Conclusion:(1) with the development of hepatic fibrosis ,the expressions of TGFβ1 in the spleen and liver of the rats increased.(2) The serum levels of TGFβ1 of the rats after splenectomy were significantly lower than which of the rats without splenectomy,and the expressions of TGFβ1 in the liver of the rats with splenectomy were less than which of the rats without splenectomy,we may infer that to some extent , splenectomy can delay the development of hepatic fibrosis by reducing the production of TGFβ1.
