Objective: To elucidate the effect of CSE on oxidative injury andapoptosis of endothelial cells,and whether transfection ofAdenovirus-mediated MnSOD to endothelial cells could partly inhibitedthe effect or not.Methods: The endothelial cells(ECV304) were cultured in vitro anddivided into control group,blank plasmid group and MnSOD group aftertransfection.They were interfered by different concentrations of CSEsolution(0%,5%,10%). We collected the cells and supernatants of eachgroup which had been interfered by different concentrations of CSE for24h,and used TBA method to measure the concentrations of MDA; usedTUNEL method to measure AI.Results:1. CSE can dose-dependently and time-dependently induce theincrease of MDA and apoptosis of the endothelial cells in control group.2. CSE can dose-dependently induce the increase of MDA andapoptosis of the endothelial cells in MnSOD group. The differencebetween blank plasmid and MnSOD group is statisticallysignificant(P<0.05). Conclusion:1. CSE can induce oxidative injury and apoptosis of endothelialcells.2. Transfection of Adenovirus-mediated MnSOD to endothelial cellscould partly inhibited the effect.
