Background: In recent times, people noticed that the formation of newblood vessel and lymphatic vessel were associated with the growth,infiltration and metastasis of the malignant tumor. The tumor could obtainabundant nutritive substance from the lumen, also spread lots of tumor cell tothe lumen. These led to the persistently growth and metastasis of themalignant tumor. Lately, in some studies, some scientists had found therelationships between the expression of thymidine phosphorylase (TP) andthe density of microvessels (MVD), the prognosis, the clinical, pathologicalfeature of the tumor. The detection of TP could play an important role in thetherapy of oncogene by some studies that were about the effect of inhibitor ofTP and TP in chemotherapy in the future. Vascular endothelial growth factor(VEGF) was the most important marker in many vascular growth factors. Itcould be a marker of metabolism and metastasis of the malignant tumor.Recently, Vascular endothelial growth factor C that was its subtype had beenfound very close relation with the growth of tumorous lymph vessel. Becausein the absence of effevtive immunohistochemical markers of lymphaticendothelium in paraffin section led to the hysteresis of lymphatic growth ofthe malignant tumor. Recently, that some specific lymphatic factors which had the potential for increasing the accuracy of detection of lymphatic growthwere found. Such as monoclonal antibody D2-40. This essay researched inthe expressions of VEGF-C,TP,lymph vessel count (LVC)and each otherrelationships by immunohistochemistry S-P. So as to provide a new methodfor the diagnosis and cure of gallbladder carcinoma.Objective: The expression of VEGF-C,TP and LVC in gallbladdercarcinoma, their relationships to some clinicpatholigic characters, and thecorrelations each other were investigated..Methods: Forty-two excision specimens were made into paraffinembeded slices after the fixation of 10% neuter formalin, adopted withimmunohistochemistry S-P to inspect the expressions of VEGF-C,TP,LVCin the forty-two gallbladder carcinoma and sixteen normal gallbladder tissues.The data that was putted into SPSS11.0 statistical software was analyzed. Theanalysis was applied by rank-sum test,two-sample/group t-test andSpearman.Results: 1.The respective positive expression rates of VEGF-C,TP inthe gallbladder carcinoma were 64.3%(27/42), their score were 2.80±2.26and 2.85±1.93, respectively. The respective positive expression rates in thenormal gallbladder tissues were 6.3%(1/16), 12.5%(2/16), their score were0.69±1.08 and 0.69±1.45, respectively. There was a significantly differenceof the VEGF-C,TP in the gallbladder carcinoma tissues and the normal gallbladder tissues (P<0.01) .2.There was a significantly difference of themean of LVC in the gallbladder carcinoma tissues and the normal gallbladdertissues(12.08±3.18 VS 4.32±1.31, P<0.01).3. The expression of VEGF-C,TP and LVC were related to the pathological type, the pathological grading,infiltration and metastasis or not (P<0.05), but were not related to the age,gender (P>0.05).4. The expression of VEGF-C and LVC,TP and LVC,VEGF-C and TP presented a positive correlation (P<0.01)Conclusion: 1. The increase of expression of VEGF-C,TP in gallbladdercarcinoma showed that they were closely related to gallbladder carcinomadifferentiated degree,infiltration and metastasis. It showed that VEGF-C,TP were probably a selective marker of generation,infiltration andmetastasis in the gallbladder carcinoma, for there were a high expression ingallbladder carcinoma. 2. The mean of LVC in the gallbladder carcinomatissues higher than in the normal gallbladder tissues, and it was closelyrelated to the pathological grading,infiltration and metastasis of gallbladdercarcinoma. LVC could be an important marker to appraise the potential ofmetastasis in the gallbladder carcinoma. 3. The expression of VEGF-C,TPand LVC presented a positive correlation. It cued that VEGF-C and TP wouldpromote the growth of tumorous lymph vessel.
