Objective The present study was designed to investigate the expression and content of VEGF and its receptor KDR in colorectal carcinoma, and observe and contrast the effect of Anti-VEGF antibody and Anti-KDR antibody on the growth of human umbilical vein endothelial cells and colorectal cancer cells LoVo in vitro, so as to confirm which is the better target for tumor vascular targeting therapy, the ligand VEGF or its receptor. Methods SABC immunohistochemical method was used to detect the expression of VEGF and KDR in 80 cases of colorectal carcinoma , Computer Image Analysis System and neighbor section quantitative analysis technique was used to contrast the content of VEGF and the content of KDR in situ . then Anti-VEGF antibody and Anti-KDR antibody were added at various concentrations to the culture medium respectively, the activity of cultured cells were measured by MTT assay. Results ?VEGF was mainly localized to the cytoplasm and the membrane of the tumor cells , and KDR was mainly localized in the membrane of vascular endothelial cells in colorectal carcinoma tissues. the positive rate of VEGF and KDR in colorectal carcinoma was significantly higher than that in normal colorectal tissue ( P<0.01 ), and they were associated with the differentiation and Dukes grade of carcinoma ( P< 0.05). ?The result of neighbor section quantitative analysis the content of VEGF were 19.2?.5 , the content of KDR were 10.5?.1, with statistically significant difference between them ( P < 0.01 ) . ? Anti-VEGF antibody had significantly inhibitory effect on colorectal cancer cells LoVo (P<0.0l) , at the same time, it had some inhibitory effect on human umbilical vein endothelial cells (P> 0.05) Anti-KDR antibody had significantly inhibitory effect on both LoVo cells and human umbilical vein endothelial cells (P<0.01) . the addition of Anti-KDR antibody had a more significantly inhibitory effect compared with the addition of Anti-VEGF antibody under the same concentration(P
