To Investigate The Effects Of NOR₁ On The Expression Profile Of Hepatoma Cell Line HepG2 Using DNA Microarray

Objective:1 To establish the stable transfection of hepatoma cell linepcDNA3.1(+)/NOR₁-HepG2.2 To investigate the effects of NOR₁ on the expression profile ofhepatoma cell line HepG2 by DNA microarray.3 To discuss the possible regulation mechinasim between NOR₁ andup-regulated/down-regulated genes.Methods:1 The plasmid pcDNA3.1(+) and eukaryotic expression vectorpcDNA3.1(+)-NOR₁ were introduced into HepG2 by liposome,respectively. The positive clones were identified by RT-PCR.2 High-throughout DNA microarray was used to screen the effectsof NOR₁ on the expression profile of hepatoma cell line HepG2.Results:1 The stable transfected cell line pcDNA3.1(+)/NOR₁-HepG2 wasestablished successfully.2 The results of DNA microarray suggested that the changes of theexpression profile induced by NOR₁ transfection were very wide.Theup-regulated and down-regulated genes were amount to 59 and 103,respectively.The most important altered genes belonged to functionalcategories of tumor, cell apoptosis,cell cycle,enzyme regulator activity, cellsignal transducer activity, cell translation regulator activity, cell transporteractivity and so on.Conclusions:1 The stable transfected cell lines pcDNA3.1(+)/NOR₁-HepG2 wasestablished. 2 DNA microarray was used to screen the effects of NOR₁ on theexpression profile of hepatoma cell line HepG2.Verified results suggestedthe results of DNA microarray were reliable.3 The influence of NOR₁ on the expression profile of hepatoma cellline HepG2 was wide.4 Some altered genes may belong to many important signalpathways. The results suggested NOR₁ may play an important role inhepatocarcinogesis.
