Objective: To evaluate the therapeutic effects of EpirubicinPolybutylcyanoacrylate nanoparticles against transplanted malignantliver tumor of rats via vena caudalis.Methods: Sixty rat models of transplanted liver were established. Aftertwo weeks, 12 rat models were selected in them randomly and werecarried out through a midline abdominal incision. The diameter of tumorwas measured. The other rat models were medially divided into fourgroups at random. Each group was administered via vena caudalis byturns.1 .Normal Saline given via vena caudalis.2. Polybutylcyanoacrylate nanoparticles given via vena caudalis.3. Free epirubicin given via vena caudalis.4. Epirubicin Polybutylcyanoacrylate nanoparticles given via vena caudalis.The survival days of the animals after administration were record .After one weeks, Abdominal exposure was carried out through a midline abdominal incision again. Volume of tumor, tumor growth rate, degree of tumor necrosis and increasing longer survival (ILS) were noted and investigated.Results: Before therapy, the mean volume of tumor was 0.28±0.09(cm~3).After therapy, statistic difference was found in volume of tumors of each group (p<0.05). Tumors of Normal Saline treated group were increased markedly, and the same as Polybutylcyanoacrylate nanoparticles treated group(p>0.05). Tumors of free epirubicin treated group and Epirubicin Polybutylcyanoacrylate nanoparticles treated group were decreased, especially in Epirubicin Polybutylcyanoacrylate nanoparticles treated group. (P<0.01). Conspicuous statistic difference was found in tumor growth rate of Epirubicin Polybutylcyanoacrylate nanoparticles treated group (p<0.01).Hyperplasia of tumor cells in Normal saline treated group was activated , cellular necrosis and degeneration was found little in the center of tumor .And the same as Polybutylcyanoacrylate nanoparticles treated group(p>0.05).Epirubicin Polybutylcyanoacrylate nanoparticles treated group was found remarkable necrosis extensively comparing with the other three groups (P<0.01).The tumor was replaced of fibril tissue and non-structure tissue. Survived time of Normal Saline treated group was 13. 33±4. 76days, Polybutylcyanoacrylate nanoparticles treated group was 14. 67±5. 39days. Epirubicin treated group was 23. 67±5. 68 days, and Epirubicin Polybutylcyanoacrylate nanoparticles treated group was 42. 83±9. 63days. With statistical analysis, the survival rate of animals of each group was analyzed with increasing longer survival. The results showed that the survival time was not prolonged between Normal Saline treated group and Polybutylcyanoacrylate nanoparticles treated group (p>0.05); Comparing with Normal Saline treated group, Survived time of epirubicin and Epirubicin Polybutylcyanoacrylatenanoparticles treated group were prolonged especially in Epirubicin Polybutylcyanoacrylate nanoparticles treated group. It demonstrated significant statistics difference between epirubicin and Epirubicin Polybutylcyanoacrylate nanoparticles treated group . (P<0.01)Conclusion: There were no difference between normal saline treated group and Polybutylcyanoacrylate nanoparticles treated group in tumor growth rate and increasing longer survival (p>0.05). Comparing with normal saline treated group and free epirubicin treated group, the tumor growth rate was all decreased significantly and increasing longer survival was all increased significantly in Epirubicin Polybutylcyanoacrylate nanoparticles treated group (P<0.01).
