| Objective: Observe the expression of inducible nitric oxide synthase (iNOS) in every pathological stage of experimental allergic encephalo-myelitis (EAE) rat.Explore the pathogenesis of iNOS of EAE rat and the therapeutic effects and mechanism of aminoguanidine to EAE rat.Methods: The EAE rat model was istablished with the homogenate of GP's spinal cord .The female Wistar rats were randomizated into five groups: control group (10 rats),NS group (10 rats),natural course of diease group(M group,30 rats),low dose Aminoguanidine treatment group(AG1 group,30 rats),high dose aminoguanidine treatment group(AG2 group,30 rats). Each of the natural course of diease group and AG1 groups and AG2 group were divided into initial stage group,peak stage group, paracme group by the neurofunction score. Infiltration of inflammatory cell was observed by HE staining method of lumbar intumescentia of the rat. The iNOS expression was measured with the immunohistochemistry method ,and the process of myelin and neuraxis was observed with electron-microscope.Results: 1 disease rate: There is no ill rats in control group and NS group .The rate is 93.3% in natural course of diease group .The latent period of the disease onseting was delayed in the AG group; The rate of AG1 and AG2 groups was lower than that of the natural course of diease group (P<0.01). Comparing with the AG1 group,the rate of AG2 group is lower (P<0.05) .2 Ethology appraisal: The neurofuction score of each AG groups is lower than that of the natural course of diease group at peaking stage and paracme (P<0.05) , and that of peaking stage of AG2 group is lower comparing with the AG1 group (P<0.05) .3 Neuropathological findings: There is no inflammatory cell infiltration of the control group and NS group, but the inflammatory cell was found in the spinal cord of the quaque stage group of the natural course diease group and AG group. The number of vasomuff consisted of inflammatory cell of AG group is fewer than that of the natural course group (P<0.05) . that of the AG2 group is lower than the AG1 group(P<0.05) .The degree of myelinolysis of the natural course diease group was more severe than that of the AG group by the electron microscope.4 iNOS expression: There is no iNOS cells in spinal cord of the control group and NS group;but the iNOS cell can be seen in the spinal cord of the natural course diease group and AG group, and the number of iNOS cell of AG group is fewer than that of the natural course diease group (P<0.05) , that of the AG2 group is lower than the AG1 group(P<0.05) .Conclusion: 1. There is the expression of iNOS cell in the spinal cord of EAE rats,and the number of iNOS cell can be concerned with the disease reactiveness.2. Aminoguanidine could reduce the incidence of the EAE rats, and relieve the neurofunction impairment and infiltration of inflammatory cell .there is a positive correlation between the dose and therapeutic effect of aminoguanidine. |
