Objective:To study the effect of anti-histamine treatment on the immune function in chronic HBV hepatitis by investigating the change of observed indexes in plasma or in serum after anti-histamine treatment.Methods:45 patients with chronic hepatitis B were collected and devided into 2 groups at random according to the order:15 patients for control group and 30 patients for treatment group, at the same time,15 healthy individuals were selected as normal control group,theatment group were treated with ketotifen based on control group,peripheral blood were collected before and after treatment,the plasma ET,HA,ALT,TBIL and HBVDNA levels,and serum IL-2,IL-4,IL-10,IL-12,IFN-γlevels were detected.Results:1.Compared to normal control group,the levels of plasma ET and HA were obviously increased in all patients before treatment,after treatment,the decreased breadth of plasma ET and HA concentration were more evident in treatment group than in control group,there was obvious statistics difference between them.2.Compared to normal control group,the levels of serum IL-2,IL-12 and IFN-γwere obviously decreased,while the levels of serum IL-4 and IL-10 were increased obviously,the ratio of Th1/Th2 was decreased significantly in all patients before treatment,after treatment,the increased breadth of serum IL-2,IL-12 and IFN-γconcentration,the decreased breadth of serum IL-4 and IL-10 concentration and the increased breadth of the ratio of Th1/Th2 were more evident in treatment group than in control group,there was obvious statistics difference between them.3.Compared to normal control group,the levels of serum ALT,TBIL and HBVDNA were obviously increased in all patients before treatment,after treatment,the decreased breadth of serum ALT,TBIL but not HBVDNA concentration were more evident in treatment group than in control group,there was obvious statistics difference between them.4.According to plasma ET concentration of normal control group,patients in treatment group were divided into two groups:ET positive and negative.After anti-histamine treatment,the decreased breadth of plasma ET,HA,serum IL-4,IL-10 concentration were more evident in ET positive group than in ET negative group,The increased breadth of serum IL-2,IL- 12,IFN-γconcentration and the ratio of Th1/Th2 were more evident in ET positive group than in ET negative group,there was obvious statistics difference between them. 5.The correlation anaysis showed that in total patients,the level of HA was positively correlated with IL-4 and IL-10,while it were negatively correlated with IL-2 and Th1/Th2 ratio respectively.Conclusion:The higher level of HA was positively correlated with Th1/Th2 unbalance in patients with chronic hepatitis B,antihistamine treatment may lighten HA,correct Th1/Th2 unbalance and improve liver function,this will provide a new idea for clinical therapy to chronic hepatitis B.
