The Expression Of Connexin40 And Connexin43 In Adult Heart

Objective:To investigate chamber-related differences in connexin40 and connexin43 protein expression in the adult heart.Analyzing the expression relationship between the two protein and finding out the linkage to the arrhythmia.To provide morphology basis for the clinical treatment and drug development.Method:Human myocardium was collected from donor.Samples of left and right atrial,left and right ventricles,interatrial septum and interventricular septum and so on were obtained from ten completely healthy donor.All the tissues were processed in paraform about 20-24 hours,the density of paraform is four percent.Paraffin imbedding as routine.Sect serial sections and choose four serial sections in every 10 slices.Thickness of the section is 6μm.Carry out the staining as follows:(1)HE staining;(2)Masson staining;(3)The glass slides were processed by polylysine,immunohistochemistry staining.Making use of HE staining and Masson staining to identify histological position.The distribution of Cx40 and Cx43 were studied immunohistochemically using of SABC method with antibody Cx40 and Cx43.At the same time we use image analysis system(Image-pro plus 5.0)to make analysis and comparison about Cx40 and Cx43 expression in chambers.All the data were disposed by statistical sofeware(SPSS 11.5).We emphasis on the different expression between left and right atrial,left and right ventricle,interatrial and interventricular,last but not least,atrial and ventricle.At last,by using linear correlation analysis to identify the expression relationship between two proteins.Result:(1)Observe HE staining under light microscope,we see the cardiocyte is stiated short columnar cell.Every cell include one elliptical nucelus,locates in the middle of it.There are two nucleus now and then.The nucleus is polymorph.Muscle fiber in the ventricle is pachy and long,but in the atrial is fine and short.Ventricular muscle fiber have branches and atrial have no. Intercalary disc were stained salmon pink,the distribution is manifest.(2)Observe Masson staining under light microscope,we see elastic fiber were stained blue.Cytoplasm,cardiac muscle fibers and red blood cell in the lumina were stained red.Nucelus were blue and brown. We can observe intercalary disc as well,stained light blue.(3)The connexin43 immunolabelling was abundant throughout the myocardium of the 4 adult heart chambers.Connexin43 granules distributed irregularly in cell side surface and cytoplasm as well as intercalated disc in adult atrial.Cx43 immunolabelling of adult ventricular myocardium was typically confined to the site of intercalated disk.The results of image analyzer showed that the amount of cormexin43 expression was higher in atrium than that of the ventricle in the adult heart.(4)The connexin40 immunolabellingwas different throughout the myocardium of the 4 adult heartchambers.The results of image analyzer showed that the amount of connexin40 expression was higher in atrium than that of the ventricle in the adult heart.The amount of Cx40 expression was higher in right atrium than that of left atrium.There is no significant difference between left and right ventricle.(5)Connexin40 and connexin43 were also expressed in interatrial,interventricular, crista terminalis,papillary muscle and so on.Connexin40 and connexin43 granules distributed irregularly in cell side surface in these positions.There is no significant difference between interatrial and interventricular.Conclusion:(1)Yhere are different expression patterns about connexin protein in adult heart. Connexin40 and connexin43 were expressed in four chambers and in atrial myocardium is more than that of in ventricular myocardium.But the amount of Cx40 expression was higher in right atrium than that of left atrium.(2)Cormexin40 and connexin43 granules distributed irregularly in cell side surface and cytoplasm as well as intercalated disc in adult atrial.And in ventricule myocardium they were typically confined to the site of intercalated disk.The expression of the two proteins showed positive correlation.
