Objective:Find a new way to preparation of composite bioactive porous MSC/Freeze-drying bone scaffold.Then,observe the result that the new type repairs the rabbit radial bone defect,the biodegradation cycle and the contribution to the osteanagenesis such as bone aiming,conduction,induction.Provide a new idea and method in tissue engineering.Methods:1.Adop the Polymeric sponge impregnation process and the modified method to prepare the composite bioactive porous MSC/Freeze-drying bone scaffold.2.choose 32 rabbits' radial bone for the bone defect models.the defect is 15mm,then embedding the support,the rabbits are randomly classified into four groups,group A is the experiment,embedding the composite bioactive porous scaffold,group B.C are the control group,embedding Freeze-drying bone and autogenous bone individually,group is the blank,embedding nothing.After operation, the animals are separately sacrificed in the forth,eighth,twelfth week.two rabbits are sacrificed in every group,then,carry out the following examination:functional assessment,X-ray examination,scanning electron microscope,histology observation,vitodynamics detection.Results:1.The composite bioactive porous scaffold's antilinear,interval porosity,and crushing strength is individually 150~300μm,62.59%~79.36%and 27MPa~53MPa 2. Histology observation and X--ray examination in the forth,eighth,twelfth week show that about the degree of the bone defect reparation,there is no significant difference between the experiment group and the autogenous bone group.Compare with the group B,the significant difference was detected.(P<0.05)In the blank,the defect is full of the fibrous tissue and the muscular tissues.Vitodynamics detection show in the twelfth week that the cmshing strength has no significant difference between the experiment group and the autogenous bone group.In the turn round examination,the experiment group has significant effect than the group C.Conclusion:Composite bioactive porous MSC/Freeze-drying bone scaffold,conductivity, degradation and induces the bone formation high performance.Implants in body has no reject reaction and allergic reaction.So it is the effective bone defect reparation material,also has its superiority as the bone tissue engineering porous scaffold.
