Objective:To investigate the proliferation function of gamma delta T(γδT)cells in peripheral blood by IL-2 and nitrogen containing-bisphosphonates(N-BP_s)-stimulated on the patients with renal carcinoma.Methods:(1)TheγδT cells proliferation assay determined T cells absolute value,γδT cells absolute value andγδT cells relative magnitude in peripheral blood of 35 patients with renal carcinoma without any treatment and of 42 healthy individuals by Flow cytometry.(2)We made a proliferation assay in vitro on the 35 patients with renal carcinoma by IL-2 and N-BP_s -stimulated before operation.26 positive patients in vitro were trial group in which we determined T cells absolute value,γδT cells absolute value andγδT cells relative magnitude after operation.(3)In trial group,we used the immunotherapy of proliferation ofγδT cells by IL-2 and N-BP_s- stimulated in vivo.Then we determined T cells absolute value,γδT cells absolute value andγδT cells relative magnitude in peripheral blood before and after the immunotherapy,taking the 42 healthy individuals as control group.Results:(1)Comparing between the patients with renal carcinoma and the healthy individuals the absolute count of all T cells was shown p>0.05,γδT cells absolute value and the percent ofγδT cells in patients with renal carcinoma were lower than in healthy individuals P<0.001.(2)In the 35 patients with renal carcinoma,the positive reaction rate ofγδT cells proliferation in peripheral blood by IL-2 and N-BP_s-stimulated in vitro was 74.28%(26/35). Comparing T cells absolute value,γδT cells absolute value andγδT cells relative magnitude in peripheral blood before and after operation,the result was shown p>0.05.(3)In trial group,the T cells absolute value,γδT cells absolute value andγδT cells relative magnitude in peripheral blood increased after the immunotherapy of proliferation ofγδT cells by IL-2 and N-BP_s -stimulated in vivo,p<0.01;then compared with control group,p>0.05,the positive reaction rate of the proliferation assay in vivo was 96.15%(25/26),higher than in vitro.(4)Consistency checking of theγδT cells proliferation trial in peripheral blood between in vivo and in vitro was shown p<0.05.Conclusion:γδT cells counting in peripheral blood of the patients with renal carcinoma is lower than healthy individuals.The proliferation function ofγδT cells in peripheral blood by IL-2 and N-BP_s -stimulated is different in variety individuality.The function ofγδT cells proliferation is defective in the patients with renal carcinoma.γδT cells counting in peripheral blood proliferates significiently by IL-2 and N-BP_s-stimulated in vivo among the patients who were positive in vitro.γδT cells proliferation assay in vitro can be regarded as a indication index for using IL-2 and N-BP_s in vivo.
