Objective:To investigate the effects of diphosphonate on proliferation and the expression of human telomerase reverse transcriptase ( hTERT) in non-small cell lung cancer cell line A549.Methods:The proliferation of A549 was assessed by Methyl thiazolyl tetrazolium( MTT) method .The morphological change of A549 treated by Ibandronic acid was observed under microscope. Cell cycle was analyzed by flow cytometry. The expression of hTERT was assessed by RT-PCR.Result:Diphosphonate could inhibit the proliferation of lung cancer cells in vitro in dose and time-dependent manner. After treated with different concentration, the cells in control group adhere well, cells were transparent, meanwhile, the cells in test groups adhere bad, the floating cells increase gradually, act as advanced stage of apoptosis. The cell cycle was arrested in G0/G1 phase. When the drug concentration was 20μmol/L, G0/G1 phase ratio achieve the peak. The expression of hTERT on A549 cells decreased with the Ibandronic acid increasing.Conclusions:Ibandronic acid can inhibit the proliferation of A549 cells in vitro. Causing G0/G1 arrest and down—regulation of hTERT appear to be the mechanism in the growth inhibition of Ibandronic acid-treated A549 cell line.
