The Proliferative Effect Of Recombinant Human Basic Fibroblast Growth Factor On HacaT Cells

Objietive To master the cell culture technology and understand the growth character of HacaT cells.To investigate the proliferative effect of rh-bFGF under different concentration and the abnormal differentiation effect of rh-bFGF under certain concentration on HacaT cells.Methods Interfer in the HacaT cells in vitro culture of rh-bFGF under different concentration and detect the cell's activity with the method of MTT.Select the cell's under the best concentration of rh-bFGF and detect its DNA ploid with FCM.Results①Observe HacaT cell's process of growth:HacaT cell adherence on the whole after passage and others are get rid of in the process of changing culture liqud.About 4-5 day's the cell growth to 80 percent of the culture flaslc and continue passage.The cell distribution uniformity and growth with sliver and fusiform.②The proliferation effect of rh-bFGF under different concentration on HacaT cell:rh-bFGF in concentration of Ong/ml,1ng/ml,5ng/ml, 10ng/ml,50ng/ml,100ng/ml showed a significant proliferative effect as compared with each other,P＜0.05.The OD is 0.137±012,0.216±0.022,0.318±0.019,0.362±0.011,0.452±0.017, 0.510±0.012.The powerful concentration is 100ng/ml.But 500ng/ml wasn't the proliferative effect as compared with the groupe of 0ng/ml,P＞0.05,The OD is 0.140±0.057③Detect the cell's DNA ploid with FCM:with FCM,rh-bFGF in concentration of 100ng/ml showed a normal poliferative effect.Conclusion rh-bFGF has significant effects on the proliferative of HacaT cells.The best response occurred when the concentration reached to 100ng/ml.We should pay attention to the importance of effective concentration,which can promote the wound healing,reduce the cicatricle,cut down reoperation,elevate the quality of patient life,short time of hospitalization, ruduce medical cost.
