Objective To compare the pharmacodynamics of seven alcoholic extractives(Fructus Tsaoko,FT;Poria,PO;Rhizoma Anemarrhenae,RA;Semen Ziziphi Spinosne,SZS;Radix Polygalae,RP;Rhizoma Chuanxiong,RC;White Pepper,WP)on anticonvulsion,and explore their mechanism of anticonvulsant action.Methods 1.Maximal electroshock seizure(MES)was used to analyse the relative of dose and effect,relative of time and effect,maximal efficacy and potency of these alcoholic extractives in mice given by ig administration.2.Metrazol seizure test(MET)was used to compare seizure latency,anticonvulsant rate and mortality of mice with every extractives.3.Pharmacodynamic parameters of alcoholic extractives were assayed by means of Pharmacological Effective Method.4.Using the model of seizure rats induced by penicillin localized injected in cortex,we studied the effect of extractives on seizure latency, frequency of epileptiform discharge,highest wave of hippocampus EEG(Recoded by RM6240C multichannel physiological signal collection and analysis recorde),the changes of convulsant behaviors and the latency of seizure.5.The neurotransmitters content related to epilepsy in brain were determined by high performance liquid chromatography(HPLC)in order to compare the effect of extractives on Glu,Asp,Gly and GABA of hippocampus.Results and conclusions: (1)All the seven extractions could dose-dependently antagonize MES in mice.Maximum efficacy of extractions ranged from maximum to minimum were RA>PO>RC>WP,RP,SZS>FT,and the sequence of potency were RP,RC,PO,SZS,RA,WP,FT.(2)Except for SZS,all the other six extractions could prolong latent period on MET in mice,and the sequence from maximum to minimum were RC,FT,RA,PO,RP,WP,SZS.And the effect of RC was better than that of LTG.(3)Except for WP produced after 45 min,the effect of other extractions produced after 30min ig,and peaked after about 1h on average,however,persistent time of them was different.Persistent time of PO was the longest one,it was 7h.RP,RC was 6h,RA,WP was 5h,and SZS was 4h,but that of FT only lasted for about 3h.It could be conjectured that all of the seven alcoholic extractives belong to short-half period preparations studied by Pharmacological Effect Method.(4)RC,PO and RA were more effective extraction to antagonize the penicillin-induced seizure.Anticonvulsant properties of them resembled LTG, FT and WP also had effect in penicillin-kindling model,but RP and SZS hadn't.(5)Through the neurotransmitters content related to epilepsy in brain determined,we can conclude blow:the anticonvulsant mechanism of PO and RC was probably due to decreasing the level of excitable amino acids Asp and Glu,however,WP was related to increasing rejection capable amino acids GABA level.The anticonvulsant mechanism of FT and RA might have relationship with the two mentioned aspects.While SZSand RP had no obvious effect on neurotransmitters content related to epilepsy in hippocampus.
