Objective: To observe the effects of Yiqiqiangxin Decoction (YQQXD) on the functional changes and mechanism for preserving heart, preventing myocardium from remodeling, improving cardiac contractility through the experiments in vivo and vitro.Methods:1 Effects of YQQXD on heart function of rats with acute cardiac insufficiency1.1 The effects of YQQXD on hemodynamics of rats with acute cardiac insufficiency40 adult male SD rats were randomly divided into 4 groups: normal control group (NC group), model control group (MC group), low-dose YQQXD group (L-YQQXD group), high-dose YQQXD group (H-YQQXD group). L-YQQXD group and H-YQQXD group were orally administered once a day for 6 days. The other 2 groups were given the same amount of saline in the same way. Then doxorubicin hydrochloride (10 mg/kg) was given intraperitoneally once to induce acute cardiac insufficiency. The MS4000U-1C biological signal collecting system was used to record and analyze the hemodynamics parameters of left ventricle after 24 hours. 1.2 The effects of YQQXD on cardiac pathological histology of rats with acute cardiac insufficiencyThe division of groups and medication were the same to above. The some middle cardiac tissue of left ventricule was cut up, and fixed in 10% formaldehyde, then sliced up, dyed by HE and Masson methods, and changes of cardiac pathological histology were observed by microscope.1.3 The effects of YQQXD on contents of SOD and MDA in serum of rats with acute cardiac insufficiencyThe division of groups and medication were the same to above. Head was cut off to obtain blood and examine the contents of SOD and MDA in serum by xanthine oxidase and TBA methods.2 Effects of YQQXD on heart function of rats with chronic cardiac insufficiency2.1 The effects of YQQXD on hemodynamics of rats with chronic cardiac insufficiency48 adult male SD rats were randomly divided into 4 groups, namely: normal control group (NC group), model control group (MC group), low-dose YQQXD group (L-YQQXD group), high-dose YQQXD group (H-YQQXD group). L-YQQXD group and H-YQQXD group were orally administered through once a day for 4 weeks after abdominal aorta banded 4 weeks. The other 2 groups were given the same amount of saline in the same way. The MS4000U-1C biological signal collecting system was used to record and analyze the hemodynamics parameters of left ventricle in rats after abdominal aorta banded 8 weeks.2.2 The effects of YQQXD on cardiac hypertrophy and cardiac pathological histology of rats with chronic cardiac insufficiencyThe division of groups and medication were same to above. After head was cut off to obtain heart from chest, the left ventricule (including ventricule septum) was divided along ventricular septum. Left ventricule (including ventricule septum) was weighted, then counted left ventricular weight/body weight (LVW/BW, mg/g), namely left ventricular weight index (LVMI), was measured. Then some middle cardiac tissue of left ventricule was cut up, and fixed in 10% formaldehyde, sliced up, dyed by HE and Masson methods, changes of cardiac pathological histology were observed by microscope.2.3 The effects of YQQXD on contents of Angâ…¡and ALD in plasma of rats with chronic cardiac insufficiencyThe methods of experiments were the same to above. Head was cut off to obtain blood and examine the contents of Angâ…¡and ALD by radiation-immunization method. 3 The effects of YQQXD on contractility of isolated atrium and papillary muscleThe hearts were isolated from mouse, SD rat, guinea-pig and rabbit, then the atria and papillary muscles were separate to observe the effects of YQQXD on contractility of atrium and papillary muscle. The MS4000U-1C biological signal collecting system was used to record and analyze the changes of spontaneous beating and contractility.Results:1 Effects of YQQXD on heart function of rats with acute cardiac insufficiency1.1 The effects of YQQXD on hemodynamics of rats with acute cardiac insufficiencyCompared with NC group, the HR, LVSP, lv+dp/dtmax of the MC group were lower, the LVEDP, lv-dp/dtmax of the MC group were higher, which had a significant difference; compared with MC group, the HR, LVSP, lv+dp/dtmax of each treatment group were higher, the LVEDP, lv-dp/dtmax of each treatment group were lower, which had a significant difference.1.2 The effects of YQQXD on cardiac pathological histology of rats with acute cardiac insufficiencyObservision by microscope: the myofibrilla in the NC group appeared normal without disruption, the nucleuses were thin and long; compared with NC group, myofibrilla disarry was observed in the MC group, obvious degeneration and a lot of inflammation cells accompanied; compared with MC group, the myofibrilla was observed in the L-YQQXD group, a few inflammation cells and disruption could be seen; The H-YQQXD group is similar to the NC group.1.3 The effects of YQQXD on contents of SOD and MDA in serum of rats with acute cardiac insufficiency Compared with NC group, the SOD of the MC group was decreased, the MDA of the MC group was increased, which had a significant difference; compared with MC group, the SOD of H-YQQXD group was increased, the MDA of the MC group was decreased, which had a significant difference; compared with MC group, the SOD of L-YQQXD group did not increased, but the MDA decreased, which had a significant difference.2 Effects of YQQXD on heart function of rats with chronic cardiac insufficiency2.1 The effects of YQQXD on hemodynamics of rats with chronic cardiac insufficiencyCompared with NC group, the LVSP and lv+dp/dtmax of the MC group were lower, the HR, LVEDP and lv-dp/dtmax of the MC group were higher, which had a significant difference; compared with MC group, the LVSP and lv+dp/dtmax of each treatment group were higher, the HR, LVEDP and lv-dp/dtmax of each treatment group were lower, which had a significant difference.2.2 The effects of YQQXD on cardiac hypertrophy and cardiac pathological histology of rats with chronic cardiac insufficiencyCompared with NC group, the LVMI, MD and CVP of MC group were higher; compared with MC group, those of each treatment group were significantly differenences.Observision by microscope: the myofibrilla in the NC group appeared normal without disruption, the nucleuses were thin and long, few collagen could be seen; compared with NC group, myofibrilla disarry and hypertrophy were observed in the MC group, obviously cellular edema and a lot of collagen accompanied; compared with MC group, the myofibrilla in L-YQQXD group, some collagen and myofibrilla hypertrophy could be seen; the H-YQQXD group is similar to the NC group.2.3 The effects of YQQXD on contents of Angâ…¡and ALD in plasma of rats with chronic cardiac insufficiencyCompared with NC group, the Angâ…¡and ALD of the MC group increased, which had a significant difference; compared with MC group, those of each treatment group decreased, which had a significant difference.3 The effects of YQQXD on contractility of isolated atrium and papillary muscle3.1 The effects of YQQXD on spontaneous beating and contractility of different isolated atriaYQQXD produced the positive inotropic effect and the negative chronotropic effect, but their effects were changed on different atria.3.2 The effects of YQQXD on spontaneous beating and contractility of isolated atria of rats with cardiac insufficiencyYQQXD produced the positive inotropic effect and the negative chronotropic effect in the concentration-dependence mannars.3.3 The effects of YQQXD on contractility of isolated rabbit papillary muscleYQQXD had the positive inotropic effect, and appeared the accumulated concentration-response curves.Conclusion:1. YQQXD can improve the changes of cardiac pathological histology of rats with doxorubicin hydrochloride-induced acute cardiac insufficiency, which may partly depend on regulating oxyradical in body and preserving cardiac myofibrilla.2. YQQXD has beneficial effects on cardiomuscular structural remodeling and prevent (delay) the development of chronic cardiac insufficiency of rats with abdominal aorta banding-induced chronic cardiac insufficiency, which may partly depend on decreasing the contents of Angâ…¡and ALD in plasma.3. YQQXD produces the positive inotropic and the negative chronotropic effects in the concentration-dependence mannars, and they show a wide range of species-dependent variation. These effects may partially contribute to improve hemodynamics of rats with cardiac insufficiency...
