Objectives:After the tumor and cardiovascular disease, diabetes mellitus has become thethird chronic non-infectious disease. Now it is an increasingly serious commonalitysanitation problem, and fearfully threat health of human. Mostly, type 2 diabetesmellitus occurs in adult, which is a major type in diabetes mellitus. Type 2 diabetesmellitus is a common genetic disease, which has complex factor and complexpolygene. It has obvious heterogeneity and its heredity and pathogenesis also are verycomplex. Circumstance instance and inheritance factors can reciprocity, which causediabetes mellitus. As an inflammatory cytokine, E-selectin can be expressed in stoke,coronary artery disease, atherosclerotic, rheumatoid arthritis, scleroderma, sepsis anddiabetes mellitus. It has great sense to learn about the E-selectin and its genepolymorphism, especially in screen high danger crowd, control start factor ininflammation and prevent related disease. It is helpful for provides the new angle ofview in open out genetics pathogenesis in diabetes mellitus to exam E-selectin geneA561C polymorphism and discuss the relationship between its polymorphism andtype 2 diabetes mellitus, and at the same time it can provide new mentality andmethod for the treatment of type 2 diabetes mellitus.Methods:1. To collect the blood samples of diabetes mellitus patients (112 case) and control subjects (140 case).2. To measure the stature, avoirdupois and blood pressure of the cases.3. To collect the blood preparation of the cases, at the same time to detect data,such as liver function, kidney function, blood serum glucose level and bloodserum lipide level.4. To extract and to purify the genomic DNA from the collected blood samples.5. Using PCR-RFLP technique, the E-selectin gene A561C polymorphism sitewas screened in 112 diabetes mellitus patients and 140 controls.6. To analyze clinical data.Results:After heredity balanced state examines for mutation of E-selectin gene A561C, itis consistent with Hard-Weinberg rule. So, we can analyze the gene frequency in thesecases. 12 (10.71%) with the heterozygote mutation (AC) were found in 112 DMpatients, and the C allele frequency is 5.36%. 4 (2.86%) with the heterozygotemutation were found in 140 controls, and the C allele frequency is 1.43%. There is nohomozygote mutation (CC) in the two groups. There has statistics differences betweentwo groups in the A allele frequency (P>0.05). In DM group and control group, SBP,BMI, TC, TG,LDL, FPG, P2PG and FINS has statistics difference (P<0.05), DMgroup is higher than the control group. Look at from the genotype, AC genotype cartieris higher than CC genotype carrier in FPG, P2PG and FINS. There has statistics difference(P<0.05)Conclusions:In the Hart nationality population in Tianjin of China, A561C genepolymorphism of E-selectin has exists. Look at from this study, we can take this siteas one of the sign for the happen of the type 2 diabetes mellitus. At the same time, wefind that the mutation participates in some metabolism target adjustment. ACgenotype carrier is higher than CC genotype carrier in FPG; P2PG and FINS. It indicates that this mutation not only exists but also plays an important role in theoccurrence and development of diabetes mellitus.
