Relationship between platelet membrane glycoproteins andparameter of platelet in.acute cerebral thrombosis[Background] Platelet activation play a crucial role in the course ofdevelopment of atherosclerosis. Activated platelet induce the inflammatory signalstransmission, contributing to thrombus formation after plaque rupture. In the processof this, platelet become activated and expose fibrinogen-binding sites on themembrane-activated glycoprotein GPⅡb/Ⅲa complex, inducing platelet aggregationand consolidate activation; release platelet a-granule membrane protein-140,eventually leading to thrombus formation.[Objective] To investigate the association of platelet membrane glycoproteinGPⅡb/Ⅲa,GMP-140,GPⅡb,GPⅢa with parameter of platelet in acute cerebralthrombosis(ACT), estimating accurately the function of platelet in order to findconvenient, econormic, reliable detection of platelet activation.[Methods] We conducted a case-control study of 51 hospital cases with ACTand randomly selected 29 cases of healthy control subjects without evident cerebralarterial disease(sex, age suited). To detect the positive percentage of activatedGPⅡb/Ⅲa complex, GMP-140,GPⅡb,GPⅢa with flow cytometer (FCM) in allpatients. Parameter of platelet were measured in all patients using automatic bloodanalyzer.[Results] 1. Comparing the data with the clinical control: The positivepercentage of activated GPⅡb/Ⅲa,GMP-140,GPⅡb,GPⅢa and MPV weresignificantly higher in the patients ; There was no difference beween the two group inplatelet count (PC),platelet distribution width (PDW) and large platelet rate (LPR).2. Hypertension, diabetes mellitus and past stroke influence the positive percentage ofactivated GPⅡb/Ⅲa; Hypertension influence the positive percentage of GMP-140(p<0.05). 3. The expression of platelet membrane glycoprotein were no significant indifferent infarction location and stroke severity of the patients group. 4. ActivatedGPⅡb/Ⅲa correlated with MPV and LPR (r=0.322, p=0.004; r=0.224, p=0.046 respectively). GMP-140 is correlated with MPV similarly (r=0.240, p=0.032). Onthe other hand there were no relationship between platelet membrane glycoprotein andPC,PDW. 5. Logistic regression analysis revealed that hypertension, activatedGPⅡb/Ⅲa were independent risk factor of ACT.[Conelusion] acute cerebral thrombosis depend on platelet activation.Platelet membrane glycoprotein and platelet parameter played along with this courseall together. Activated GPⅡb/Ⅲa correlated with MPV significantly. Hypertension,activated GPⅡb/Ⅲa were independent risk factor of ACT. The detection of Plateletmembrane glycoprotein and platelet parameter can estimate accurately the functionof platelet. They provided a evidence for clinic prevention, diagnosis, therapy andprognosis estimation in acute ischaemic cerebral disease.
