| Obsjective: To investigate the effects of Tirofiban on actived-platelet and prethrombotic state in rabbit model of iliac artery injury by balloon angioplasty.Methods: 40 New Zealand white rabbits, mean weight: 2.4±0.3kg(range: 2.1~2.8kg), were randomized to 3 groups: 11 for pre-treatment with Tirofiban(Pre-T, n=11), 13 for Tirofiban-treatment(T, n=13), 6 for placebo-control(C, n=6).All rabbits were lavaged with aspirin(12mg/kg) and clopidogrel(16mg/kg) 8~12hs before experiment, then abrosiaed (Tirofiban was injected sustainingly with a minipump right after being lavaged in Pre-T untill the end of experiment). Anesthetize with 3% pentobarbital sodium, dissociate the right femoral artery carefully, put in a 4F catheter quickly, tight with a surgery thread, put in a PTCA balloon(2.5mm in diameter) along the catheter, expand the balloon under 10 atmospheric pressure slowly, push and pull the balloon for 30 seconds, then stop for 1 mimute, repeat the above step twice, Tirofiban or placebo was injected right after the model was built. Put another catheter in the right carotid artery in the same way. 100U/Kg heparin was given just after the catheter had been put in and 50U/Kg/h followed.Referent standard of sP-selectin, TXB2, 6-Keto-PGF1α, von Willebrand factor was made before experiment from 17 of the 40 rabbits. Two samples of blood were drawn before and after experiment: one for packing fraction of platelet, the other for sP-selectin, TXB2, 6-Keto-PGF1α, von Willebrand factor.Monitor the fluctuation of iliac systolic blood pressure(SBP) and diastolic blood pressure(DBP), document the time when they reach to meet. Some models were taken arteriography. The injuried iliac and the counter-part were taken out and made into common HE pathological section to analyze the character and composition of the thrombus after the experiment.
