Objective To study the constituents from Sorbaria sorbifolia and the inhibition of5, 2', 4'-trihydroxy-6, 7, 5'-trimethoxyflavone on proliferation hepatoma HepG-2 cells.Methods The constituents from Sorbaria sorbifolia chloroform extract wereisolated with sillia gel column chromatography and identified by physicochemicalproperties and spectroscopic analysis. MTT assay was used to examine the effect of 5, 2',4'-trihydroxy-6, 7, 5'-trimethoxyflavone on the proliferation of HepG-2 cell and flowcytometry was used for the cell cycle distribution. Apoptosis of HepG-2 cells wasinvestigated further by means of inverted microscope, HE staining.Results Five compounds were isolated and identified as 5, 2', 4'-trihydroxy-6, 7,5'-trimethoxyflavone (â… ), 5, 7-dihydroxy-8-methoxyflavone (â…¡), daucosterol (â…¢),benzoate (â…£) and p-hydroxybenzoic acid (â…¤). 5, 2', 4'-trihydroxy-6, 7, 5'-trimethoxyflavone inhibited the proliferation of HepG-2 cell and the effects were in atime-and-concentration dependent manner by MTT assay. 5, 2', 4'-trihydroxy-6, 7, 5'-trimethoxyflavone can also induced apoptosis, the G2/M phase was arrested and theratio of apoptosis there was significantly difference than that of control group,respectively.Conclusion Five compounds were isolated from Sorbaria sorbifolia chloroformextract for the first time. 5, 2', 4'-trihydroxy-6, 7, 5'-trimethoxyflavone inhibited theproliferation of HepG-2 cell and the effects were in a time-and-concentration dependentmanner by MTT assay. 5, 2', 4'-trihydroxy-6, 7, 5'-trimethoxyflavone can also inducedapoptosis, the G2/M phase was arrested and the ratio of apoptosis there was significantlydifference than that of control group, respectively.
