Effects of Chinese angelica alcohol extracts on skeletal muscles contraction and its preliminary mechanismOBJECTIVE: To investigate the effect of Chinese angelica on antagonizing fatigue and its preliminary biochemical mechanism in rats and mice.METHODS: Male Wistar rats, weight 180-200g and male KunMing mouse, weight 20-22g. The wistar rats used for the measurement of the skeleton muscles contraction experiment and biochemical indicator, the mouse useds for a measurement to force a swimming time. The 28 experiment mouse were randomly divided into 4 groups by the habituation swimming screening and training: control group, Chinese angelica low group, Chinese angelica middle group, Chinese angelica large group, with seven in each. Affuse stomach with Chinese angelica alcohol extracts (low dose 750mg/kg,middle dose 1500mg/kg,large dose 3000mg/kg, drug concentration low dose 25%,middle dose 50%,large dose 100%, control group with 0.9%Nacl 32 ml/kg) 2 weeks. Intragastric administration for an hour on 14th day, carry out skeleton muscles contraction experiment and measurement of weight loading swimming times in mouse. Twenty eight rats were also randomly divided into 4 groups( I. e. control group, low dose group, middle dose group, large dose group), with seven in each. affuse stomach with Chinese angelica Oral liquid(low dose 350mg/kg,middle dose 750mg/kg,large dose 1400mg/kg, control group with tales doses 0.9%Nacl) 2 weeks. Intragastric administration for an hour on 14th day, carry out skeleton muscles contraction experiment and to make use of measurement of biochemical indicator.The experiment make use of RM-6240 system,observed the effect of electrical stimulate sciatic nerve on the contractility of gastrocnemius and soleus muscle, recorded latent period of single twitch of skeletal muscles, contraction extent, 1/2 relaxation time of single twitch, contraction extent of tetanus, latent period and fatigue index number. The assaying of Superoxide Dismutase (SOD), Glutathione Peroxidase (GSH——PX), Lactic Acid (LD) and Malondialdehyde(MDA) were carried out.RESULTS: 1 The swimming time and weight indicator of mice.①Chinese angelica alcohol extracts intragastric administration 14 days later, the control group average force swimming times of mice 180.17±26.35s, but the administration of Chinese angelica extracts groups sigfiificant prolonged the swimming time of mice, the large dose group 720.23±31.48s, the middle dose group 540.70±38.17s, the low dose group 300.45±21.81s. There are the obvious dose-effect relationship. P<0.05②Chinese angelica alcohol extracts also increase the weight of mice, the weight of the administration of Chinese angelica extracts groups compared with the control group's, average increased 2.69±0.45g, P<0.01. 2 Contraction of the skeletal muscle: (1) single twitch indicator:.①Gastrocnemius: the large dose group and low dose group of Chinese angelica compared with control group, shorten latent period, extend 1/2 relaxation time of single twitch, P<0.05; large dose group, increased the maximal contraction extent of single twitch, of gastrocnemius, P<0.05.②Soleus muscle: each groups of Chinese angelica compared with the control group, shorten latent period, extend 1/2 relaxation time of single twitch, P<0.05; in addition, except the low dose group, the maximal contraction extent significant changes were observed, other groups obviously increase the maximal contraction extent of single twitch of soleus muscle, P<0.05. (2) Tetanus contraction①Gastrocnemius: the large dose group and middle dose group compared with the control group, increase the maximal contraction extent of tetanus, shorten latent period, decrease the percentage of fatigue index number, P<0.05; low dose group except for obviously decreasing the percentage of fatigue index number of tetanus, other indicators no significant changes were observed, P<0.05.②Soleus muscle: the large dose group and middle dose group compared with the control group, increase the maximal contraction extent of tetanus, shorten latent period, decrease the percentage of fatigue index number, P<0.05; low dose group, the latent period, the maximal contraction extent of tetanus and the percentage of fatigue index number no significant changes were observed, P>0.05, don't statistics to learn difference. 3 Biochemical indicator: gastrocnemius and soleus muscle the large dose group biochemical indicator: compared with the control group, increase MDA,GSH-PX,SOD and decrease LD, P<0.05.CONCLUSION: The Chinese angelica alcohol extracts can improve antagonizing fatigue of the skeleton muscles of rats; and have a certain dose dependent. This effect may be pass to lower the free radicals in the skeleton muscle and decrease the effect of the lactic acid in the skeleton muscles.
