Two-photon microscopy has attracted high interests on investigation of biomedicine recently, because of its several outstanding characteristics, including intrinsic optical section, lower photon-bleaching and photo-damaging, high contrast and longer penetrating distance. In this thesis, a novel two-photon microscopy combined multichannel imaging mode with spectral imaging mode was developed to investigate biological tissues, including human dermis, rabbit arterial wall, and mouse oral mucosa. The experimental results indicate that two-photon microscopy combined multichannel imaging mode with spectral imaging mode is a potential tool to quantitatively visualize and analyze intrinsic species of tissues, and it can also be used to investigate metabolism of cells and analyze chromatin structure. The technique may prove useful to clinical diagnostics as well as to basic biological research.
