Collagen fiber is an important part of human tissue. The change of microstructure in collagen fiber is closely related to Skin aging, wound healing, corneal disease, liver fibrosis, osteoarthritis and malignant tumor. This paper is aim at taking high-contrast imaging of collagen fiber in beast, stomach and pancreas based on multi-photon microscopy. Because of multiplicity and complexity of biological tissue, the intensity of excited of different organizations are different. When the light interacting with matter, the physical mechanism of second harmonic generation(SHG) and two-photon excitation fluorescence(TPEF) are different and the intensity of excited of them is also different when they are optimal results. We obtain the spectral information of collagen fiber at different intensity of excited and diagram of relation between the intensity of signal and power of excited laser. First, we obtain the SHG image of collagen fiber and take down the power of laser. Then we obtain the power of excited of TPEF. Our research can obtain the SHG and TPEF high-contrast image of collagen fiber faster and more accurately.
