Objective To investigate the expression of a disintegrin and metalIoprotease 8(ADAM8) in human serum and tissue with non-small cell lung cancer, to evaluate thecorrelation between ADAM8 expression and clinical and pathological feature of NSCLCand explore value of ADAM8 in diagnosis and therapy of NSCLC.Methods (1)Serum level of ADAM8 was detected in 50 patients with lung cancer,15 patients with benign pulmonary disease and 15 cases in health group by ELISAmethod, serum level of CEA was detected too by RIA method in all groups. (2)Expression of ADAM8 was detected in all 49 cases of lung cancer, 28 cases ofparacancer tissue and 13 cases with benign pulmonary disease by tissue microarray andimmunoradiometric assay, expression of EGFR was also detected in all groups.Results (1)The serum levels of ADAM8 and CEA in group of NSCLC were higherthan those in groups with benign pulmonary disease and health(P<0.05), but there wereno significant difference between benign pulmonary disease and healthgroup(P>0.05).(2)The serum levels of ADAM8 statistically correlated with farmetastases and TNM stage(P<0.05), but not with sex, age,smoking history andpathological kinds (P>0.05) in NSCLC group.(3) The serum levels of CEA statisticallycorrelated with pathological kinds, far metastases and TNM stage(P<0.05), but not withsex, age and smoking history (P>0.05) in NSCLC group. (4) The diagnosticsensitivity(%)of ADAM8 and CEA for NSCLC was 84% and 66%,statisticallysignificance among them (P<0.05); the diagnostic specificity(%)of ADAM8 and CEAfor NSCLC was 80% and 84%, no statistically significance among them (P>0.05). Areaunder the ROC curve of ADAM8 and CEA was 0.864 and 0.813.(5) The paralledcombined testing of ADAM8 and CEA increased the sensitivity to 94% for NSCLC,and serial combined testing increased the diagnostic specificity to 86.7%.(6)The expressionof ADAM8 and CEA was both higher in NSCLC group than that in groups with benignpulmonary disease and normal tissue(P<0.01), but no significant correlation was detectedin benign pulmonary disease and normal tissue group(P>0.05). (7)Expression of ADAMand EGFR statistically correlated with TNM stage and lymph node matastases(P<0.05) in NSCLC, not with pathological kinds(P>0.05), but there was statisticallysignificant correlation between the differentiation level and the latter (P<0.05). (8)Therewas accordant expression of ADAM8 and EGFR in NSCLC tissue, and value of kappawas 0.522; positive correlation was detected between them(r=0.5888).Conclusion (1) ADAM8 and CEA were over-expressed in NSCLC tissue,andaccordant among them.(2) The overexpression of ADAM8 and EGFR may play animportant role in invasion and matastasis in NSCLC.(3) The serum levels of ADAM8 canbe used for diagnosis in NSCLC,and its diagnostic value was superior to CEA.(4)Combined detection of serum ADAM8 and CEA can signicantly improve the diagnosticsensitivity and specificity in NSCLC.(5) Regulated function of EGFR ligand and othercytokines, ADAM8 can be new target in treatment of NSCLC.
