Objective:To investigate the abnormality of Her-2 protein and gene in NSCLC, and identify the association betweeen clinicopathological features, prognosis and Her-2 aberrations amongst protein and gene.Methods:140 cases of NSCLC tissues(89 squamous cell carcinoma,51 adenocarcinoma) were selected to The experiment, while 70 normal tissues as matched group.(1)Immunohistochemical method was used to detect the protein expression of TTF-1、Napsin A、CK5/6、 p63 and Her-2.(2) Her-2 genetic alterations were detected using fluorescence in situ hybridization(Fl SH).(3) To analyse the relationship betweeen clinicopathological features, prognosis and Her-2 aberrations amongst protein and gene.Results:1. Her-2 overexpression was detected in 0 case(0%) with the total number of 70 normal tissues,and 17(12.14%) cases with the total number of 140 cases which was obviously higher than normal tissues(P<0.05).2. Her-2 overexpression was associated with the pathological type of NSCLC,which was detected more frequently in adenocarcinoma. with a significant difference(c2=4.19,P=0.04).3. 40 cases presence Her-2 gene copy number gain(28.57%), and Her-2 gene amplification was detected in 6 cases(4.29%), while Her-2 gene multicopy was detected in34 cases(24.29%). Her-2 overexpression was associated with Her-2 gene amplification(P<0. 01),while no relationship between Her-2 overexpression and Her-2 gene multicopy(P>0.05).4. Her-2 gene amplification is associated with the pathological type(P=0.024),smoking(P=0.048) and younger age(P=0.015). None clinicopathological features presence correlation with Her-2 gene multicopy(P>0.05).5. There was no significantly difference in survival between patients without and with Her-2 overexpression and Her-2 gene abnormality(P>0.05).Conclusions:1. The rate of Her-2 overexpression in NSCLC is obviouslyis high than in Normal tissue.2. Her-2 overexpression is related to the pathological type of NSCLC,which was detected more frequently in adenocarcinoma squamous cell carcinoma.3.Her-2 gene amplification occurred most frequently in patients who never smoke, of younger age and who had ADC histology,The difference was statistically significant.None clinicopathological features presence correlation with Her-2 gene multicopy.4.Her-2 overexpression was associated with Her-2 gene amplification but Her-2 gene multicopy.5.No statistical difference in survival was observed comparing patients with or without abnormality of Her-2 protein and gene.
