The Effects Of Sodium Ferulate On The Renal Interstitial Fibrosis In Rat Kidney With Unilateral Ureteral Obstruction

Objective:To observe the expression of bFGF and ET-1 in rat kidney with unilateral ureteral obstruction in the development of renal tubulointerstitial fibrosis .Method:Forty five male Sprague-Dawley rats were equally divided into three groups randomly: sham operation group, UUO group and sodium ferulate (SF)-treated group. The rats in UUO group and sodium ferulate (SF)-treated group were subjected to right ureter ligation, the rats in the UUO-A group were received sodium ferulate intraperitoneally daily in the dosage of 150mg/Kg , and sodium chloride in tales doses in SOR and UUO groups. Rats were sacrificed after 14 days, 21 days and 28 days of uretcral obstruction(UUO)in batch. The obstructed kidney was taken out for histopathologic examination, urea nitrogen and creatinine 24 hours urinary protein excretion were also observed. The expression of bFGF and ET-1 were analyzed with immunohistochemical examination.Result: The results of UUO rat model experiment in vivo showed:①The obstructed kidneys is pelviectasis, renal cortex thinningz, and the size is large. Hematoxylin and eosin staining showed that interstitial fibrosis of obstructed kidneys was observed obviously after UUO compared with SOR and UUO-A group.②The urea nitrogen and creatinine increased significantly in the rats of of UUO group, but with the time of UUO, the opposite side kidney compensate gradually, and the urea nitrogen and creatinine decreased to a certain extent. The urea nitrogen and creatinine of UUO-A group decreased significantly compared with UUO-A group, which showed that sodium ferulate will ameliorate renal lesion. There was no significance of 24 hours urinary protein excretion between the three groups at each time.③The expression of bFGF, ET-1 in the tubulointerstitum were increased in UUO group, and the semi-quantitative values were also much higher than the SOR group and UUO–A group . There is a correlation between the expression of bFGF, ET-1 and the degree of tubulointerstitial fibrosis. The expression of bFGF and ET-1 in is much lower than the UUO–A group.Conclusion:The degree of tubulointerstitial fibrosis may be much more severe with the time of UUO in both groups, Sodium ferulate has obvious ameliorating effect on renal tubulointerstitial fibrosis in rats with UUO.
