Monopril In Combination With Diovan Ameliorate Renal Interstitial Fibrosis In Rat Kidney With Unilateral Ureteral Obstruction

Objective:To investigate Monopril in combination with Diovan ameliorate renal Interstitial fibrosis in rat kidney with unilateral ureteral obstruction(UUO), To observe the expression of bFGF in rat kidney with unilateral ureteral obstruction in the development of renal tubulointerstitial fibrosis.Method:sixty eight male SD rats were equally divided into three groups randomly:sham operation group(n=20),UUO group (n=24) and treatment group (n=24).The rats in UUO group and treatment group were subjected to right ureter ligation, At different time points of treated for 1,2,3,4 weeks, we measured protein-uria in 24 hours of rats, blood urea nitrogen, serum creatinine, The first two groups lavage with 1ml distilled water, sooner or later each one times, the treatment group the morning Monopril (0.5mg/100 g), the afternoon Diovan (4mg/100 g). Rats were sacrificed after 1,2,3,4 weeks, The expression of bFGF was analyzed with immunohistochemical examination.Result:The results of UUO rat model experiment in vivo showed The obstructed kidneys is pelviectasis, renal cortex thinningz, and the size is large. Hematoxylin and eosin staining showed that interstitial fibrosis of obstructed kidneys was observed obviously after UUO compared with SOR and UUO-A group. The renal pathological results by HE staining had showed that focal inflammatory cells infiltrated into renal tissues at1th week, mesangial proliferated lightly pile monocyts/moscrophages infiltrated into tubulointerstitial regions, Thetubule epithelial cells swelled.proteinuria casts in tubule were observed at 2th week. With prolongation in experiment time, these pathological changes aggravate gradually.At 4thweek had showed that glomerular capillaries were collapsed,mesangial proliferated and its matrix to broad,tubule were dilatalted or atrophied widespread,heavy proteinuria casts presented in tubular cavities,the mass fibrin accumulated in tubulointerstitial. But those pathological changes were ameliorated significantly in treated groups.There was no significance of 24 hours urinary protein excretion between the three groups at each time.I The urea nitrogen and creatinine increased significantly in the rats of UUO group,but with the time of UUO, the opposite side kidney compensate gradually,and the urea nitrogen and creatinine decreased to a certain extent, in comparison with shame operation group, bFGF was only slightly expressed in the tubular epithelial cells and non-expressed in the glomerulus in sham group rats. bFGF expression was increased notably in tubular epithelial cells at 4thweek in untreated group,and increased gradually with the progression of disease(P<0.05).the expression of bFGF of UUO and treatment group was markedly increased (P<0.05). In comparison with UUO group, treatment group significantly decreased the expression of bFGF (P<0.05).Conclusion:Combined with Monopril and Diovan can decrease the expression of bfgf. Suggesting it can delay the aggravation of renal tubulointerstitial fibrosis.
