Objective To observe the protection on the lung from ischemic reperfusion injury in the rabbits modle ,we preconditioning the group with sodium ferulate (SF) before I/R,and then studay the principles.Methods The eighteen rabbits were divided into three groups randomly :Control group (n=6,the left lung hilus were clamped to ischemic and then reperfusion as I/R);Sham group (n=6 ,thoractomy was performed without clamping left lung hilus and SF therapy);Preconditioning group (SF)(n=6,treated with 100mg/kg SF before I/R ).All the rabbits were tracheotomy and then intubated for the assistance of respirator .Following the thoractomy ,we performed the I/R model in this way: occluding the left lung hilum for 60 minites and then reperfusing for 120 minites. We collected blood from the common carotid with intubating at the time points (before ischemia,ischemia 60 minites,reperfused 30 minites,reperfused 120 minites.), centrifugated the blood for serum to detect Superoxide dismutise (SOD )and Nitric oxide(NO).The left lungs were discarded to determined the wet/dry ratio W/D,the tissue content myeloperoxidase (MPO ).The lung tissues histopathological changes and the lung tissue damage were determined in the microscope. nuclear factor (NF)-κB were assayed by immnuhistochemitry.Results After ischemia(60 minutes) and reperfusion(120minutes),the test indexes of the groups are different obviously.In the Sodium Ferulate(SF) group, the histopathology present that the less destroy alveoluses, alveoluses hemorrhage and Inflammatory cell infiltration by light microscope than the I/R group); Lung damage degree(LTD) presenting the lower red cell and Inflammatory cell infiltration than the I/R(Ischemia-Reperfusion) group (P<0.01)is not different obviously from the S(Sham) group.The serum NO and SOD are improved in the SF group . the difference between SF group and I/R group is expressly (P<0.01)but not with the S group and SF group(P>0.05). In I/R group , nuclear factor (NF) –Κb and from lung tissue aremore activated and expressed in the nucleus than S group and SF group,they present the Buffy, the gray scale are significant deviation from S group and SF group(P<0.01). Myeloperoxidase(MPO) present the same result as the above factors.Conclusions The classical lung I/R model occluding the hilus and then reperfusing in vivo leading to lung ischemic reperfusion injury undoubtedly .The effect reach its peak at the time point 30 minite and 2 hour reperfused.The aggregated neutrophil(Polymorphonuclear PMN) and Sequestrated to lung, more inborned oxyradical (OFR)and reduced endogenous NO are composed of the factor to LIRI mainly. More importantly, the activated nuclear factor (NF)-Κb play the trig role in vacious cycle of LIRI . Sodium Ferulate(SF) has protect the lung from suffering IRL evidently.The Preservated fact may inhibit the polymorphonuclear neutrophils (Polymorphonuclear PMN) aggregating to lung injury, suppressing NF-κB , decrease the OFR and increase the content of endogenous NO and SOD.
