Objective: To study the protective effect of pharmacological precondition induced by sodium ferulate (SF) on isolated rat rnyocardium and its machanism. Methods: Isolated SD rat hearts were divided into 5 groups at random: Normal group (hearts were perfused by oxygenated perfusate for 100 minutes); Ischemia/Reperfusion group (hearts suffered from 40 minute global ischemia/30inute reperfusion); Ischemia precondition group (hearts were preconditioned by three times 5inute global ischemia/5inute reperfusion before it suffered from 40inute global ischemia/30 minute reperfusion); SF group (previously hearts were perfused with oxygenated perfusate administered with 1.69mmol/L SF then subjected to 40inute global ischemia/30inute reperfusion); Gliberclamide group (previously hearts were perfused with oxygenated perfusate administered with 1.69mmol/L SF and 30itmol/L Glib before it suffered from 40inute global ischeniia/30inute reperfusion). Results: Compared with hR group, 1.69mmol/L SF precondition obviously increased coronary flow, improved heart function; notably promoted activity of superoxide dismutase (SOD) and glutathion peroxidase (GSHX), reduced malondialdehyde (MDA) content; alleviated calcium overload in myocardial cell; lessened the incidence and severity of ventricular arrhythmias; furthered activity of Na TPase, Mg2 TPase and Ca2 TPase in myocardium. These protective effects were attenuated by 3Omoh/L Gliberclamide administered. Conclusions: 1. SF precondition can improve heart function and resist arrhythmia. 2. SF precondition can resist toxicity of free radical and Ca2 verload. 3. The protective effect of SF precondition is related with the opening of ATP ensitive potassium (KATP) channels.
