Objective To study the growth inhibiting and apoptosis induced by Aspirin on the proliferation and apoptosis of the human colorectal cell line Lovo.also to explore its possible mechanisms of anticancer effects.Methods In vitro.exponential phase human colorectal cancer cell line Lovo was treated by aspirin in various concentration(s100μg /ml,200μg /ml,300μg /ml ) .cell morphology changes was observed by optical microscopy.using MTT assay to detect absorbance value and calculate inhibition or survivial rate.and then the effect of cell growth inhibition was assayed by MTT method.the induction of cell cycle and apoptosis by aspirin was assessed by flow cytometry, the expression of Survivin,Bcl-2 and Bax protein were analyzed by immunocytochemistry(ICC). the expression of Survivin mRNA was determined by reverse transcriplase polymerase chain reaction(RT-PCR) to study the machanism of apoptosis induced by aspirin.Results MTT assay show that Aspirin could inhibit the growth of the human colorectal carcinoma cell line Lovo, and the inhibitory effect was dose-and-time dependent (P<0.05). the inhibition rate was the maximum under Aspirin dentisy was 300μg/ml which was 56.9%.There was an apoptotic peak in Lovo cells after incubating with aspirin detected by flow cytometry.Quantitative analysis result from DNA bargragh was that cell apoptotic ratio of 100μg/ml,200μg/ml,300μg/ml density was 13.5%,21.3%, 48.3%.the difference was significant compared to the control group,s3.2%.the cells in G1/G0 stage multed as the density of Aspirin increased, cells in S stage descended gradually,while cells in G2/M stage have no variation.Also the expressions of Survivin and bcl-2 were downregulated,there were many eumorphism in cell in control group.however the expression of bax was upregulated significantly as the density of Aspirin increased.many eumorphism could be seen in cell membrane and emdochylema rather than control group. the expression of Survivin mRNA was determined by reverse transcriplase polymerase chain reaction(RT-PCR). Expression of Survivin mRNA was downregulated through ladder strap as the inceasing of Aspirin concentration.Conclusion Cell crowth inhabition and induction of apoptosis could be carried out by Aspirin in a dose-and-time dependent manner, and its possible mechanism was downregulating the expressions of Survivin and Bcl-2. upregulating the expression of Bax.
