Objective To investigete the inhibition effect of 125IUdR onthe proferation of the human bladder cancer cell line Sca-BER and theinfluence of 125IUdR on the cell-cycle and cell apoptosis.Methods: 125IUdR were added to the RPMI1640 culturing medium. Theamount of 125IUdR uptake by Sca-BER cells was evaluated throughmeasuring the radioactivity per cell; the killing effects of 125IUdR onSca-BER cells were estimated by growth-curve, colony forming method. Todetermine the co-inhibition effects of MTX, 125IUdR and MTX were addedto medium, measuring the radioactivity per cell. The influence of 125IUdR onthe cell-cycle and the cell apoptosis was explored by flow-cytometry(FCM)and DNA gel elctrophoresis technique.Resuits:①The amounts of 125IUdR uptake by Sca-BER were growing upas a function of 125IUdR dose in the medium(r=0.99). The surviving fractionwas correlated with the concentration of 125IUdR(r=-0.9). The LD50 of125IUdR group was (1.17±0.27) kBq/mL. Sca-BER cells were efficientlyinhibited by 125IUdR in a time-and-concentration-dependent mannar, whileNa125 I showed little inhibition effect on the proferation of Sca-BER cells; Inthe growth curve experiment, the cell double time was prolonged; In thecolony forming experiment, a shoulder-free survival curve was yielded;125IUdR and MTX co-inhibited on the Sca-BER cells.②The cell cycleanalysis showed 125IUdR made the S phase Sca-BER cells decrease,compared with the control-group cells, p<0.01; Ap could be seen in the 37kBq/mL 125IUdR-treated Sca-BER cells by FCM; 37 kBq/mL 125IUdR-treated Sca-BER cells was analyzed by DNA gel electrophoresis, revealed atypical ladder form of DNA fragments, and 370 kBq/mL 125UdR- treatedSca-BER cells showed no regular pattern of banding.Conclusions: The human bladder cancer Sca-BER cell line can be inhibitedby 125IUdR, the Auger electron of 125IUdR have the DNA-targetedirradiation. 125IUdR has potential for the treatment of bladder cancer.
