Objective: To investigate the protective effects of prostaglandin A1(PGA1) on ischemia-reperfusion during rabbit lung-transplant, and explore its possible mechanism.Methods: Thirty-two rabbits were randomly divided into tow groups: control group and trail group. In control group, the harvested lung blocks were flushed with and stored in the modified low-postasssium-dextron(LPD) solution at 4℃for 4h. In trail group the harvested lung blocks were flushed with and stored in the modified LPD solution containing PGA1(80ng/L) at 4℃for 4h. After preservation, the rabbit lung-transplant ischemia-reperfusion models were established and the lungs were perfused for 1h. PaO2 were measured at just beginning time, 15min, 30min, 45min, 60min time during reperfusion. After reperfusion, the lung tissue wet-to-dry(W/D) ratio and myeloperoxidase(MPO) activity were obtained. The protein expression of nuclear factor-kappa B(NF-κB) and inter-cellular adhesion molecule-1(ICAM-1)in the lung tissue was also detected by using immunohistochemistry and Enzyme-Linked Immunosorbnent Assay(ELISA)at the end of reperfusion. Light microscopic and ultrastructural changes were also examined.Results: (1)The level of PaO2 decreased gradually, the value of PaO2 in control group was lower than that in PGA1 group. The provide-lung function in PGA1 group is better than that in control group. (2)The difference of PaO2 in control group and in PGA1 group was obvious after 30min and 45min reperfusion(p<0.05), and the difference was greater after 60min reperfusion(p<0.01). (3)The value of W/D, MPO, the protein expression of NF-κB and ICAM-1 in PGA1 group decreased more evidently than in contral group (P<0.05). (4) The light microscope change of lung tissues in control group is damaged hardly than PGA1 group. (5)The transmission electron microscope demonstrated the lung tissue in control group had mild alveolar septae with congestion obviously in the micrangium. Red blood cells could be seen in the alveolar space. Some alveolar epithelial cells appeared edema or exfoliation with lamellar body evacuated and occasional areas of aqtocytolysis. However, the lung tissue in PGA1 group is better than that in control group.Conclusion: (1) This research confirmation,during rabbit lung-transplant, the modified LPD solution which containing PGA1 can decrease the activity of MPO evidently in provide-lung tissues, and inhibit the concentration of PMN, increase the super microstructural of the lung tissues. (2) PGA1 can inhibit the expression of NF-κB in provide-lung tissues after rabbit lung-transplant. (3) PGA1 can inhibit the expression of ICAM-1 in provide-lung tissues, so alleviate the ischemia-reperfusion injury, and improve the protective effect of lung preservation.
