Diabetes is a chronic disease that occurs when the pancreas does not produce enoughinsulin, or when the body cannot effectively use the insulin it produces. With the oldage trend and high living level, the incidence of diabetes is higher and higher everyyear. At present, all the treatments of diabetes have disadvantages. Insulin has shortaction, and should be injected to patients. Transplant is expensive and can be painful.Oral medications are less potent and liable to cause diarrhea, nausea and liver failure.It is urgent to exploit effective antidiabetic medicine with low toxicity. In this study,we investigated the hypoglycemic effects of the new vanadium complexes on thediabetic mice and the signaling transduction mechanism in order to provide.sustainment for further research.(1)By cell culture in vitro, the vanandium complexes were selected with methylthiazolyl tetrazolium (MTT) colorimetric assay. Results showed that vanadiumcomplexes promote the growth of the cells when the concentration is lower than 10μmol/L, but when the concentration is higher than 100μmol/L, the survival ratesdecrease obviously, and presented toxicity is less related to incubated time.(2)Established mice diabetic models induced by aUoxan. Then the diabetic mice weregavaged with vanadate complexes to see their hypoglycemic effects. Compared withraw material, NVC-T2 and NVC-T3 decreased the glucose levels more efficiently.The morphology of tissue showed vanadium complex NVC-T2 did not have toxicity.(3)Expressions of proteins associated with insulin receptors signaling pathway in theliver of diabetic mice cured by new vanadium complexes and in Chang liver cellstreated by new vanadium complexes were assessed by Western blot.Conclusion: Tp'and SCN ligands can reduce the toxicities of vanadium complexes.NVC-T2 has gypoglycemic effect, and it can regulate lipid metabolism. Insulin-likeeffect of this vanadium complex may be involved in PKB pathway.
