The Apoptosis Of Human Tongue Cancer Cell Line Tca8113 Induced By Oxaliplatin And The Alteration Of Survivin Gene Expression

Objective:To investigate whether the third generation of platinum antitumor drug,oxaliplatin could induce apoptosis in human tongue cancer cell line Tca8113 in vitro,and what the change of expression of inhibitory apoptosis gene,survivin in this process was.Methods: Tca8113 cells cultured in vitro were as a model.The influence of proliferation of Tca8113 cells under different oxaliplatin concentration (16,32,64,128,256μmol/L) was measured by thiazolyl blue tetrazolium bromide method(MTT),and The Inhibitory Concentration of 50%(IC50) was calculated.Then morphological changes on cellular and subcellullar structures and apoptosis cells were observed and photographed with inverted microscope and electron microscopy under the condition of IC50, alteration of cell cycle and apoptosis were analyzed by using flow cytometry(FCM).The expression of survivin were detected with immunohistochemical technique(IHC) and reverse transcription- polymerase chain reaction technique(RT-PCR),respectively.Results: 1.Concentrations of oxaliplatin from 16 to 256μmol/L, could inhibit proliferation from 24 to 96h,the effect reached its peak in 48～72h.2.Morphological changes such as chromatin condense,apoptotic bodies were observed. 3.Cell cycle analysis indicated that oxaliplatin blocked cell proliferation in S and G2/M phase,the apoptotic peak emerged when treated with oxaliplatin for 48,72 hours,and the percent of apoptosis cells apparently increased(P<0.05vs control).4.When treated with oxaliplatin for 24,48,72 hours, the Tca8113 cell expression of survivin was up-regulated in all three points by IHC(P<0.05vs control),except for in 24h point by RT-PCR(P>0.05vs control). Conclusion: 1.Oxaliplatin can inhibit the proliferation and induce apoptosis of Tca8113 cells in vitro.2.In this process, the cell expression of survivin was up-regulated,which may be one of mechanism of oxaliplatin chemotherapy resistance.
