Objective:To explore the possibility of Pantoea agglomerans lipopolysaccharide(LPSp)acting as an adjuvant of purified Vero cell vaccine for human use.Methods:Balb/c mice were randomly divided into six groups: the six groups are injected with LPSp,purified Vero cell vaccine, LPSp+purified Vero cell vaccine,rabies Ag,rabies Ag + LPSp,0.9% NaCl respectively.Each groups are divided into three small groups, each small group are designated to have the single dose or the multiple dose of immunization.The single dose inoculated at Oth day, the 2 doses inoculated at 0 and 15th day,the multiple dose inoculated at 0,7,14 and 21th day.all the preparations were immunized with 0.2ml through back hypodermia injection for each mice.The serum anti-rabies virus titers were detected by ELISA.Comparing the antibody Ig G,IgM titers in different group and observing the kinetics of IgG titer in different period.Results:It was found that the single dose immunization with the LPSp + purified Vero cell vaccine could induce and higher antibody response than the normal single dose immunization with purified Vero cell vaccinre,furthermore,2 doses of inoculation with LPSp + purified Veto cell vaccine could attain the immune efficiency of the 4 doses immunization with vaccine.The same,the single dose immunization with rabies Ag + LPSp could induce earlier and higher antibody response than the single dose immunization with rabies Ag,also the 2 doses of inoculation with rabies Ag + LPSp could attain the immune efficiency of the 4 doses immunization rabies Ag.At the same time we found when the mice have the same dose the antibody of the LPSp + purified Vero cell vaccine was higher than the antibody of the rabies Ag + LPSp.The antibody persistent time is potentially longer in additional LPSp groups.Conclusion:It can concluded that LPSp can act as an adjuvant to improve the immune efficiency of purified Vero cell rabies vaccine KEY WORDS:LPSp,Vero cell rabies,vaccine,adjuvant... |
