OBJECTIVE To explore the relationship between the expression of FAF1 in gastric cancer and its clinical parameters, through testing the expression at mRNA level and protein level and apoptosic index on the samples from human gastric cancer . METHODS: Real time polymerase chain reaction (Real time PCR, RT-PCR) was employed to determine the expression of FAF1mRNA in cancerous, paracancerous and normal tissues from 40 cases of gastric cancer, respectively. Immunohistochemistry and TUNEL were used to detect the expression of FAF1 and apoptosic index in cancerous, paracancerous and normal tissues from 40 cases of gastric cancer, respectively.RESULTS part I The expressive levels of FAF1mRNA in gastric cancer and its clinical significance. The expressive levels of FAF1mRNA in the cancerous tissues (0.27±0.12) were significantly lower than that in the paracancerous tissues (0.40±0.06) and normal tissues(0.48±0.08 p<0.01). The expression of FAF1mRNA in gastric cancer was closely correlated with cell differentiation, distant place diversion, but not correlated with the age, sex, depth of invasion, stage (TNM), lymph node mets and the tumor size.part II The expression of FAF1 in gastric cancer and its relationship with the AI and its Clinical Significance : The positive expression rates of FAF1 in the cancerous tissues (37.50%) were strikingly lower than that in the paracancerous tissues (62.50%) and normal tissues(72.50% p<0.05). The expression of FAF1 in well-differentiated cancerous tissues were much higher than that in poorly-differentiated ones (p<0.05). The apoptosic index in gastric cancer was closely correlated with cell differentiation, distant place diversion, infiltrating depth and clinical stages (p<0.05). The AI of gastric cancerous cells in positive expression for FAF1 was much higher than in the negatives (p<0.01).CONCLUSION: There were relationship between the expression of FAF1 and the gastric Carcinogenesis. The low expression in cancerous tissues may decrease the apoptosis and promote the gastric cancer development.
