Objective: To observe the expression changes of IL-12 and IFN-γin plasma by infusing different doses of free hemoglobin in hemorrhagic shock animals , then to know the FHb's effect on animals'immune function. Methods: We collected the hemorrhagic shock animals'venous blood samples: after resuscitation, 16 hours after resuscitation, and 48 hours after resuscitation. Results: The expression of IL-12 and IFN-γobviously dropped after transfusing FHb in hemorrhagic shock animals(P<0.01). The plasma levels of IL-12 and IFN-γachieved the minimum value when 16 hours after resuscitation, and they achieved the foundation value levels when 48 hours after resuscitation. Their trends were negatively correlated with the dose of FHb. In 15mg/kg group animals, the plasma level of IL-12 elevated when 24 hours after resuscitation, and maintained a high level until 48 hours after resuscitation. Conclusion: In the early stage of hemorrhagic shock, FHb significantly restrained the function of antigen-presenting cells and caused the inhibition of cellular immunity. So it lowered the hemorrhagic shock animals'resistance to pathogen and increased the probability of infection. Also the research found that as the dose of FHb increased, there would be a high inflammatory response period after resuscitation. This period might increase the damage in major organs. It might increase the incidence of MODS.
