Skin is the largest organ in the human body, not only has a physical defense function, but also has an special immune function which is close correlated to general immunity system. We will establish and authenticate Epidermal Langerhans Cell Absenced Mice (ELCAM) and RA Cutaneous Reaction Mice (RACRM) model, then followed by investigate the expression of cytokines in skin, subcutaneous muscle and blood after transdermal absorption of rHu IFN-α2b. After establish and authenticate ELCAM and RACRM model, we detect protein and mRNA expression of IFN-α, IL-18, IFN-γ, IL-10, IL-12 from the blood, skin and subcutaneous muscle after per cutem with rHu IFN-α2b cream to BALB/c mice and ELCAM and RACRM model by ELISA and Real-Time PCR. We also dectect protein and mRNA expression of IFN-α, IL-18, IFN-γ, IL-10, IL-12 from the PAM212 cell lines and L-929 cell lines with rHu IFN-α2b 6h directly or after intervention with CP and RA 3h. The results show the tissue and cell lines after administrated will change the expression of cytokines markedly. It confirm the skin immune barrier(SIB) is exist when per cutem with the biomarcomolecular drug, and SIB have effects of immunologic surveillance and immune defence.
