Objective: To observe the apoptosis effect of vitamin E succinate (VES) on the growth of androgen-independent prostate cancer cell line PC-3 in vitro. Methods: We established control and experimental groups (categorized as 25,50,75,100,125mg/L), and the latter were exposed to VES for 24h, 48h and 72h respectively. MTT method and flow-cytometry were used to detect the vitality and apoptosis rate of the PC-3 cells. Meanwhile, susceptibility test were performed to compare the efficacy between VES (25, 75mg/L) and Mitoxantrone (25, 75mg/L) respectively. Moreover, the distribution change of cell-cycle was observed after PC-3 cells were exposed to VES for 48h. Results: The vitality of experimental groups was significant lower than the control group (P<0.05), which was negative related to concentraten and action time of VES while the vitality of control group was positive related to them. The apoptosis rate of experimental groups was much higher than the control group, which shows a positive correlation with concentraten and action time of VES. Moreover, after 48h VES treatment, the proportion of the S stage PC-3 cells increased while the proportion of the G1 stage PC-3 cells decreased along with the incleased concentration of VES. And the proportion of the G2 stage PC-3 cellls didn't show much change as concentration of VES increased. However, the curve of the proportion of the S stage and G1 stage cells according to the concentration of VES showed a platform stage when VES concentration come to 75mg/L respectively. The susceptibility test showed that 25mg/L Mitoxantrone is more effective than 25mg/L VES while 75mg/L Mitoxantrone is less effective than 75mg/L VES in term of inhibition effect to PC-3 cells (P<0.05) . Conclusions: VES has anti-proliferation and apoptosis induction effect on PC-3 cells and the mechanism of the apoptosis effect is related to the blockade of cell cycle. It is indicated that the 75mg/L VES showed a best apoptosis effect after 48h treatment, cause 80% PC-3 cells apoptosis, which is more effective compare to Mitoxantrone as the same 75mg/L concetraten. Therefore, VES has a promising future for prostate cancer therapy as a chemotherapeutics, which help us to establish new therapeutic concepts for the prevention and treatment of prostate cancer.
