Objective: The effect of Vitamin E Succinate (VES) on Nasopharyngeal Caicinoma (NPC) cells has not been reported before both home and abroad. In this research, we observed the growth inhibition and apoptosis-induced effects of VES on human NPC cells. The effect of VES on CNE-2 cells was observed by inverted phase contrast microscope and transmission electron microscope. The date might provide theoretical basis for the clinical treatment of NPC.Methods:The growth inhibition of VES in different concentrations (5,10,20 and 40μg/ml) and different time (24,48 and 72h) by MTT assay; The effect of VES induced-apoptosis on CNE-2 cells was observed by inverted phase contrast microscope and transmission electron microscope; The apoptosis rate was determined using flow cytometry (FCM); Cell cycle regulated by VES was analyzed by FCM. The statistical analysis was executed by SPSS 13.0 software, using the repeated measurement analysis of vatiance (ANOVA) ,chi-square test and chi-square test division. Statistical significant level was considered as"alpha equal 0.05".Results:The effect of cytotoxicity of VES was found on CNE-2 cells. The proliferation of CNE-2 cells was inhibited by VES. The effect of growth-inhibition is correction with concerntrations and treated time. After treated by 5μg/ml VES for 24h, it was not siginficant difference compared with the controll group(P=0.085). In other groups,the satistical analysis had siginficant difference between the test groups and the control group(P<0.05). The inhibition curve shows that the effect of growth-inhibition was gradually increasing along with the increasing of concerntrations of VES and treated time. The characterized apoptosis cells induced by VES were seen under inverted microscope and transmission electron microscope. After treated by VES for 48h,the apoptotic ratio of CNE-2 Cells significiantly increased. With the increasing of concentration,the apoptotic ratio is increasing. The satistical analysis had siginficant difference(P<0.05). After treated by VES for 48h,the change of cell cycles tells us that the ratio of G1 phase is increasing with the increasing of concentration. Statistical graph shows that the changes in G1 phase was positively correlated with the concentration. The satistical analysis had siginficant difference between the test groups and the control group(P<0.05).Conclusion: In this study ,we demonstrated that VES was able to inhibit the proliferation of the CNE-2 cells,and the effect is positive correlation to the treated time and concentrations. Morphological changes of cells were seen under inverted microscope and transmission electron microscope. VES can induce apoptosis in the CNE-2 cells. VES could regulate the cell cycle of CNE-2 cells,and the proportion of the G0/G1 phase cells was increasing.
