Objective: For decades, Attention deficit hyperactivity disorder (ADHD)has been seen primarily as a most common behavior disorder affecting young children, primarily in boys. The origin of ADHD is unknown, dopamine metabolic dysfunction may be one of the main reasons. We want to investigate the correlation between the polymorphism of dopamine beta hydroxylase gene (DBH) IVS5 Taq I (rs2519152) and the ADHD of Chinese Han population in Shanghai.Methods: The DNAs of 117 children diagnosed ADHD according to the DSM-IV criteria were extracted and PCR was performed to examine the alleles (A1,A2) and genotypes (A1/A1,A1/A2,A2/A2) polymorphism of DBH IVS5 Taq I. A case-control association study and a family-based association study with Haplotype-based Haplotype Relative Risk (HHRR),Exact Transmission-disequilibrium Test (ETDT) and Chi-square test were used to analyze the correlation between the DBH IVS5 Taq I and ADHD.Results:(1) There was significant difference in genotypes (p=0.006) and alleles (p=0.001) of DBH IVS5 Taq I between 117 children with ADHD and 186 normal control group.(2)There was no significant difference in genotypes between the 76 children with ADHD and within-family control group (p=0.185). HHRR results showed that there were more A2 alleles of DBH Taq I than A1 alleles in the 76 children with ADHD (p=0.079). Exact Transmission Disequilibrium Test (ETDT) results showed no significant difference between A2 alleles and A1 alleles (p=0.136).Conclusion: There was correlation between the ADHD and DBH (rs2519152). There were more A2/A2 genotypes in the ADHD than in normal control group. A1 alleles may be protective factor, A2 alleles may be risk factor. Objective: To investigate the correlation between ABO blood type and the attention deficit hyperactivity disorder (ADHD) of Chinese Han population in Shanghai.Methods: Using glass sheet method to determine ABO blood type of 119 children with ADHD and 488 normal control group. Chi-square test was used to analyze the correlation between ABO blood type and the ADHD. The PCR-RFLP technology was used to detect the genotype of children with ADHD and their parents. A family-based correlation study with HHRR and Chi-square test were used to analyze the correlation between the ABO gene polymorphism and ADHD.Results:(1) There was no significant difference between 119 children with ADHD and 488 normal control group in ABO blood type (p=0.681).(2) There was significant difference between 18 girls with ADHD and 206 female normal control group in ABO blood type (p=0.02). There were more O,A blood type than B blood type in 18 girls with ADHD (p=0.011, p=0.019).(3) There was no significant difference between 101 boys with ADHD and 282 male normal control group in ABO blood type (p= p=0.674).(4) HHRR results showed that there wasn't significant difference between 76 children with ADHD and within-family control group in the allele of ABO (p=0.283).Conclusion: O,A blood type are risk factors in girls with ADHD, but B blood type is protective factor.
